Organismy jsou schopny přizpůsobovat své fyziologické procesy pravidelnému střídání dne a noci. U savců je tato schopnost zajištěna časovým systémem tvořeným centrálními cirkadiánními hodinami lokalizovanými v suprachiasmatických jádrech hypotalamu a periferními hodinami přítomnými v různých tkáních a orgánech. Tyto hodiny koordinují řadu fyziologických funkcí, včetně regulace metabolismu glukózy a energetické rovnováhy organismu. Cirkadiánní systém je synchronizován především vnějšími časovými signály, mezi něž patří zejména střídání světla a tmy a rytmu příjmu potravy. Pokud však dojde k narušení této synchronizace, může nastat desynchronizace mezi centrálními a periferními hodinami. Tento stav je spojován s poruchami metabolické regulace a může přispívat k rozvoji metabolických onemocnění. Tato práce se zaměřuje na charakteristiku cirkadiánního systému a jeho úlohu při regulaci metabolismu. Zvláštní pozornost je věnována periferním cirkadiánním hodinám a přehledu nejčastějších faktorů, které mohou vést k cirkadiánní desynchronizaci a jejich vlivu na vznik a rozvoj diabetu mellitus 2. typu. Organisms are able to adapt their physiological processes to the regular alternation of day and night. In mammals, this time-keeping system consists of central circadian clocks located in the suprachiasmatic nuclei of the hypothalamus and peripheral clocks present in various tissues and organs. These clocks coordinate a wide range of physiological functions, including the regulation of glucose metabolism and the energy balance of the organism. The circadian system is primarily synchronized by external time cues, particularly the light-dark cycle and the timing of food intake. However, disruption of this synchronization may lead to desynchronization between central and peripheral clocks. This condition is associated with disturbances in metabolic regulation and may contribute to the development of metabolic diseases. This thesis focuses on the characteristics of the circadian system and its role in metabolic regulation. Special attention is given to peripheral circadian clocks and to an overview of the most common factors that may lead to circadian desynchronization, as well as their potential role in the development of type 2 diabetes mellitus.
