Zuzana Klekarová Školitel: RNDr. Klára Konečná, Ph.D. Název diplomové práce: Určení antifungální aktivity vybraných, nově syntetizovaných molekul Cíle práce: V teoretické části bylo cílem přiblížit problematiku fungálních infekcí a jejich léčbu. V experimentální části práce bylo cílem určit antifungální aktivitu nově syntetizovaných látek AP2-IZ a JST-8-IMB vůči vybraným kvasinkovým kmenům a výsledky porovnat se zavedenými antifungálními léčivy. Dílčím úkolem bylo studium cidního účinku těchto látek v čase. Metody: Inhibiční koncentrace (IC50, IC90, MICVIS) byly stanoveny pomocí mikrodiluční bujónové metody. Pro určení hodnot inhibičních koncentrací (IC50, IC90) byla využita spektrofotometrická detekce. Pro studium cidního účinku v čase vůči referenčnímu kmeni kvasinky Candida albicans ATCC 90028 byla využita metoda "time-kill" eseje, přičemž trend poklesu viabilních kvasinek byl sledován v časech T=0; 3; 6; 24 hodin, následně byly stanoveny hodnoty log CFU/ml. Výsledky: Hodnoty inhibičních koncentrací IC5024h pro látku AP2-IZ se pohybovaly v rozmezí 2,13 - 34,14 mg/l a u látky JST-8-IMB 2,23 - 18,16 mg/l. The objective of the experimental section was to determine the antifungal activity of the newly synthesized compounds AP2-IZ and JST-8-IMB, against selected yeast strains and to compare the results with established antifungal agents. A secondary objective was to study the time-dependent cidal activity of these compounds. Methods: Inhibitory concentrations (IC5O, IC90, MICVIS) were determined using the broth microdilution method. Spectrophotometric detection was used to determine the inhibitory concentrations (IC50, IC90). To study the time-dependent killing effect against the reference strain of Candida albicans ATCC 90028, the "time-kill" assay method was used with the trend of the decline in viability of yeast cells monitored at time points T=0; 3; 6; 24 hours, followed by the determination of log CFU/ml values.
