[cze][eng] Apoferitin je proteinová globulární schránka složená z 24 podjednotek, které se rozpadají a zase skládají v závislosti na okolních podmínkách. Této unikátní vlastnosti se dá využít pro enkapsulaci léčiv v rámci cílené terapie nádorových onemocnění. Enkapsulací se zvýší selektivita a biologická dostupnost léčiva v nádorové buňce, což by mohlo vést k zásadnímu snížení systémových nežádoucích účinků. Tato práce si klade za cíl připravit lidský apoferitin pomocí rekombinantního expresního systému Escherichia coli a následně testovat jeho schopnost enkapsulovat vybrané cytostatikum. Exprese byla indukována pomocí IPTG a následná purifikace byla založena na vysoké termostabilitě apoferitinu, která umožňuje odstranění nežádoucích proteinů. Zakončena byla dvoukrokovým vysolováním pomocí síranu amonného. Jako modelové léčivo pro enkapsulaci byl zvolen ellipticin. Testovalo se několik typů buněk jako nejvhodnější se jeví buňky kmene BL21(DE3)pLysS s dobou indukované exprese 24 hodin.Azároveň byl do proteinové schránky produkovaného apoferitinu úspěšně enkapsulován ellipticin. Klíčová slova: apoferitin, Escherichia coli, rekombinantní exprese, nanotransportéry, ellipticin, enkapsulace.Apoferritin is a globular protein shell composed of 24 subunits that disassemble and reassemble depending on environmental conditions. This unique property can be utilized for the encapsulation of drugs within the framework of targeted therapy for cancer diseases. Encapsulation increases the selectivity and bioavailability of the drug in the tumor cell, which could lead to a fundamental reduction in systemic side effects. The aim of this work is to prepare human apoferritin using a recombinant expression system in Escherichia coli and subsequently test its ability to encapsulate a selected cytostatic agent. Expression was induced using IPTG, and the subsequent purification was based on the high thermostability of apoferritin, which allows for the removal of undesirable proteins. It was concluded with a two-step salting-out process using ammonium sulfate. Ellipticine was chosen as the model drug for encapsulation. Several cell types were tested, the BL21(DE3)pLysS strain with a 24-hour induction period proved to be the most suitable. Furthermore, ellipticine was successfully encapsulated into the protein cage of the produced apoferritin.
