[eng][cze] The mammalian immune response to viral infection relies on a coordinated effort between innate signalling, driven by the type I interferon (IFN) response, and intrinsic immunity, mediated by constitutive restriction factors. An important component at this interface is the promyelocytic leukaemia (PML) protein, the structural scaffold of PML nuclear bodies (PML NBs). PML NBs and PML protein actively participate in the cellular antiviral response. Among the human PML variants, isoforms hPMLII and hPMLIV have been shown to positively regulate the type I IFN pathway. While PML NBs are established mediators of antiviral defence, their interaction with the Polyomaviridae family represents a biological paradox: despite the restrictive potential of these structures, polyomaviruses actively localise their replication and assembly in close proximity to them. Using murine polyomavirus (MPyV) as a model system, this thesis explores the functional and spatial role of murine PML and PML NBs during viral infection. To investigate the spatial dynamics of this interplay, a CRISPR/dCas9 visualisation system was adapted to specifically target and track MPyV DNA within viral replication centres. The targeting capability and functionality of this system were successfully validated in both fixed and living cells....Imunitní odpověď savců na virovou infekci spočívá v koordinované spolupráci vrozené imunity, založené na interferonové (IFN) odpovědi typu I, a přirozené imunity, charakterizované konstitutivními restrikčními faktory. Klíčovou roli zde hraje protein promyelocytární leukémie (PML), který tvoří kostru jaderných tělísek PML. Jaderná tělíska PML a protein PML se aktivně účastní antivirové obrany. Pro dvě ze sedmi izoforem lidského proteinu PML je známo, že se účastní regulace IFN odpovědi typu I. Ačkoliv jsou PML tělíská známá jako mediátory antivirové obrany, jejich interakce s čeledí Polyomaviridae představuje biologický paradox: navzdory jejich restriktivnímu charakteru polyomaviry cílí svou replikaci a tvorbu nových virionů do blízkosti těchto struktur. Za využití myšího polyomaviru (MPyV) jako modelového systému tato práce zkoumá funkční a prostorovou roli myšího PML a jaderných domén PML při virové infekci. Aby bylo možné studovat prostorovu dynamiku této interakce, vizualizační systém CRISPR/dCas9 byl adaptovaný, aby rozpoznával DNA MPyV ve virových replikačních centrech. Funkční exprese systému byla úspěšně ověřena ve fixovaných i živých buňkách. Dále tato práce zkoumá regulační roli dvou specifických izoforem myšího PML, mPML2 a mPML3, při IFN odpovědi typu I pomocí kvantifikace exprese genu...
