[cze][eng] Gen PDX1 (pancreatic and duodenal homeobox 1) kóduje transkripční faktor PDX1, který reguluje časný vývoj pankreatu a na glukóze závislou produkci inzulinu. Homozygotní ztráta funkce tohoto genu vede k agenezi pankreatu a způsobuje neonatální diabetes a pankreatickou insuficienci, zatímco heterozygotní ztráta je spojena s neautoimunitním diabetem vzniklým v dětství či mladé dospělosti. Cílem této práce je prozkoumat vliv variant nejasného významu v genu PDX1 nalezených u dětí s neautoimunitním diabetem na transkripční aktivitu jejich produktů (mutantů PDX1), tj. na produkci inzulinu testovanou in vitro. V experimentální části byly metodou cílené mutageneze připraveny plazmidové konstrukty, jejichž správnost byla ověřena Sangerovým sekvenováním. Tyto vektory byly transfekovány do buněčné linie HEK293. Exprese mutantních proteinů byla ověřena metodou Western blot a jejich vliv na aktivitu inzulinového promotoru byl hodnocen pomocí duální luciferázové eseje. Výsledky práce ukázaly, že všech pět testovaných variant (p.Met122Val, p.Leu158Gln, p.Ile171Asn, p.Trp134Ter a c.188delC) vykazovalo signifikantně zvýšenou transkripční aktivitu ve srovnání s wild type proteinem, přičemž nejvýraznější efekt byl naměřen u varianty p.Ile171Asn. Zvýšená transkripční aktivita mutantů PDX1 je pravděpodobně obrazem...The PDX1 gene (pancreatic and duodenal homeobox 1) encodes the transcription factor PDX1, which regulates early pancreatic development and glucose-dependent insulin production. Homozygous loss of function of this gene leads to pancreatic agenesis and causes neonatal diabetes and pancreatic insufficiency, while heterozygous loss is associated with non-autoimmune diabetes developing in childhood or early adulthood. The aim of this work is to investigate the effect of variants of uncertain significance in the PDX1 gene, that were found in children with non-autoimmune diabetes, on the transcriptional activity of their products (PDX1 mutants), i.e., on insulin production tested in vitro. In the experimental part, plasmid constructs were prepared using site-directed mutagenesis, and their correctness was verified by Sanger sequencing. These vectors were transfected into the HEK293 cell line. Expression of mutant proteins was validated by Western blot analysis, and their effect on insulin promoter activity was assessed using a dual luciferase assay.
