Tato bakalářská práce se zabývá molekulárním a buněčným mechanismem stárnutí a možností jeho farmakologického ovlivnění pomocí geroprotektiv. Stárnutí je funkční úpadek způsobený postupnou ztrátou homeostázy. Geroprotektiva jsou látky, které tento úpadek zmírňují či oddalují, a chrání tak organismus před stárnutím. Mezi tyto látky se řadí například rapamycin, resveratrol, spermidin, metformin a mnohé další. Některé z těchto sloučenin fungují jako mimetika kalorické restrikce a cílí na evolučně konzervované signální dráhy. Kvasinka Saccharomyces cerevisiae je díky vysoké evoluční konzervovanosti těchto drah vhodným modelovým organismem pro pochopení molekulárního působení stávajících geroprotektiv i pro testování látek nových. Tato práce formou literární rešerše poskytuje ucelený přehled o molekulárních cílech jednotlivých geroprotektiv a popisuje jejich vliv na dobu dožití (lifespan) a dobu strávenou ve zdraví (healthspan). This bachelor thesis focuses on the molecular and cellular mechanisms of aging and the potential of its pharmacological modulation using geroprotectors. Aging is a functional decline caused by a progressive loss of homeostasis. Geroprotectors are substances that mitigate or delay this decline, thus protecting the organism from aging. These substances include, for example, rapamycin, resveratrol, spermidine, metformin, and many others. Some of these compounds act as caloric restriction mimetics and target evolutionarily conserved signaling pathways. Due to the high evolutionary conservation of these pathways, the yeast Saccharomyces cerevisiae is a suitable model organism for understanding the molecular action of existing geroprotectors, as well as for testing new compounds. Through a literature review, this thesis provides a comprehensive overview of the molecular targets of individual geroprotectors and describes their effect on lifespan and healthspan.
