[cze][eng] Tato diplomová práce se zabývá vývojem automatizovatelného průtokového systému pro elektrochemickou detekci DNA na uhlíkové pracovní elektrodě. Navazuje na předchozí výzkum v oblasti elektrochemie nukleových kyselin a průtokových analytických metod a jejím cílem je transformace klasického vsádkového uspořádání na automatizovatelný průtokový systém s důrazem na zvýšení reprodukovatelnosti, snížení manuální manipulace a zkrácení doby analýzy. V rámci práce byly testovány dvě 3D tištěné tenkovrstvé průtokové elektrochemické cely lišící se konstrukcí a způsobem dávkování vzorku. Byly optimalizovány experimentální podmínky zahrnující způsob zavádění vzorku, realizace adsorpčního kroku v režimu zastaveného toku a postupy regenerace povrchu pracovní elektrody. Jako modelový systém byla použita ssDNA analyzovaná lineární voltametrií na elektrodě z vysoce orientovaného pyrolytického grafitu. Výsledky ukazují, že přechod na průtokové uspořádání vede k významným změnám v proudové odezvě, zejména vlivem disperze vzorkové zóny a omezeného kontaktu analyzované látky s povrchem elektrody. Optimalizací experimentálních podmínek, zejména úpravou objemu dávkovaného vzorku a režimu proudění, se...This thesis focuses on the development of an automated flow system for the electrochemical detection of DNA on carbon-based electrodes. It builds on previous research in the field of nucleic acid electrochemistry and flow analytical methods, and its main objective is to transform the conventional batch arrangement into a fully automated flow system, with an emphasis on improving reproducibility, reducing manual handling, and shortening analysis time. Within this work, two 3D-printed thin-layer flow electrochemical cells differing in design and sample introduction were tested. Experimental conditions were optimized, including the mode of sample introduction, implementation of the adsorption step under stopped-flow conditions, and procedures for regeneration of the working electrode surface. Single-stranded DNA (ssDNA) was used as a model system and analyzed by linear sweep voltammetry at a highly oriented pyrolytic graphite electrode. The results demonstrate that the transition to a flow arrangement leads to significant changes in current response, mainly due to dispersion of the sample zone and limited contact between the analyte and the electrode surface.
