Polyploidní mutanti vznikají v přírodních populacích rostlin opakovaně a nezávisle v každé generaci. Mezi mechanismy jejich vzniku patří polyploidizace prostřednictvím neredukovaných (2n) gamet, somatická polyploidizace a vzácně také polyspermie. Hlavní roli při vzniku polyploidů však hraje produkce 2n gamet, ke které dochází v důsledku meiotických chyb, obvykle spočívajících ve vynechání prvního nebo druhého meiotického dělení. Tato bakalářská práce se soustředí zejména na genetické a environmentální faktory, které ovlivňují produkci 2n gamet v rostlinných populacích. Popisuje mutace v konkrétních genech vedoucí k chybám v průběhu meiózy zdokumentované na modelových rostlinách a zdůrazňuje vliv přírodních abiotických a biotických stresových faktorů, ale i antropogenního znečištění, které k vyšší frekvenci meiotických nepravidelností přispívají. Práce se věnuje i metodám detekce 2n gamet, od klasických mikroskopických pozorování tvaru a velikosti pylových zrn po průtokovou cytometrii fluorescenčně obarvených jader samčích gamet. Na závěr je diskutován potenciální evoluční význam zvýšené frekvence tvorby 2n gamet včetně jejich příspěvku k úspěšnému uchycení nových polyploidních linií a zprostředkování genového toku přes reprodukční bariéru odlišné ploidie. Klíčová slova polyploidizace, neredukované... Polyploid mutants arise repeatedly and independently in natural plant populations in every generation. The mechanisms underlying their formation include polyploidization via unreduced (2n) gametes, somatic polyploidization, and, rarely, polyspermy. However, the production of 2n gametes, which occurs as a result of meiotic errors, usually involving the omission of the first or second meiotic division, plays the primary role in the formation of polyploids. This bachelor's thesis focuses primarily on the genetic and environmental factors that influence the production of 2n gametes in plant populations. It describes mutations in specific genes leading to errors during meiosis documented in model plants and highlights the influence of natural abiotic and biotic stress factors, as well as anthropogenic pollution, which contribute to a higher frequency of meiotic irregularities. The work also addresses methods for detecting 2n gametes, ranging from classical microscopic observations of pollen grain shape and size to flow cytometry of fluorescently stained male gamete nuclei. Finally, the potential evolutionary significance of the increased frequency of 2n gamete formation is discussed, including their contribution to the successful establishment of new polyploid lines and the mediation of gene flow across...
