[cze][eng] Tato bakalářská práce se zabývá mechanismy regulace fluidity cytoplazmatické membrány u bakterií čeledi Enterobacteriaceae v reakci na environmentální stres. Cytoplazmatická membrána představuje pro gramnegativní bakterie klíčové rozhraní, jehož fyzikální stav bezprostředně podmiňuje funkci membránových proteinů, energetický metabolismus a celkovou integritu buňky. Hlavním cílem rešerše je analýza strategií homeoviskózní adaptace, které enterobakterie využívají k zachování membránové homeostázy při změnách teploty, pH, osmotického tlaku a v přítomnosti membránově aktivních látek. Text detailně popisuje molekulární dráhy vedoucí k remodelaci lipidového složení, zejména prostřednictvím změn v saturaci a délce acylových řetězců mastných kyselin a post-syntetické tvorby cyklopropanových mastných kyselin. Pozornost je věnována také regulační roli dvoukomponentových systémů a alternativních sigma faktorů, které tyto procesy koordinují. Závěrečná část práce diskutuje vliv membránové plasticity na virulenci, patogenezi a rozvoj antibiotické rezistence, přičemž poukazuje na potenciál biosyntézy membránových lipidů jako cíle pro vývoj nových antimikrobiálních látek. For Gram-negative bacteria, the cytoplasmic membrane represents a crucial interface whose physical state directly determines the function of membrane proteins, energy metabolism, and overall cellular integrity. The primary objective of this literature review is to analyze homeoviscous adaptation strategies employed by enterobacteria to maintain membrane homeostasis during changes in temperature, pH, osmotic pressure, and in the presence of membrane-active compounds. The text describes in detail the molecular pathways leading to the remodeling of lipid composition, particularly through changes in the saturation and length of fatty acid acyl chains and the post-synthetic formation of cyclopropane fatty acids. Attention is also given to the regulatory role of two-component systems and alternative sigma factors that coordinate these processes. The final section of the thesis discusses the impact of membrane plasticity on virulence, pathogenesis, and the development of antibiotic resistance, highlighting the potential of membrane lipid biosynthesis as a target for the development of novel antimicrobial agents.
