[cze][eng] Tato bakalářská práce se zabývá procesem specifikace a migrace savčích primordiálních zárodečných buněk (PGCs) a jejich následnou rolí v patogenezi germinálních nádorů. Formou literární rešerše aktuálních poznatků, založené na srovnání myšího a lidského modelu, je popsán vývoj těchto buněk od raného epiblastu až po kolonizaci genitálních lišt. Text nejprve popisuje molekulární indukci PGCs v reakci na signální dráhy BMP, WNT a Nodal, poté ustanovení germinální identity pomocí transkripčních faktorů tzv. tripartitní regulační sítě: Blimp1, Prdm14 a AP2γ. Zvláštní pozornost je věnována migraci PGCs, která zahrnuje pasivní transport v důsledku morfogeneze zadního střeva a aktivní transport regulovaný chemotaktickými signály, zejména osou CXCL12/CXCR4 a adhezivními interakcemi systému c-Kit/SCF. Práce dále shrnuje proces rozsáhlého epigenetického přeprogramování, který je pro PGCs v této fázi specifický. Závěrečná část je věnována souvislostem mezi selháním migrace a vznikem ektopických PGCs, které představují zdroj extragonadálních germinálních nádorů. Jsou diskutovány mechanismy umožňující těmto buňkám uniknout apoptóze a zahájit maligní transformaci. Klíčová slova: Primordiální zárodečné buňky (PGCs), migrace, germinální nádory, savci, ektopické PGCs, epigenetické přeprogramování.This bachelor thesis examines the process of specification and migration of mammalian primordial germ cells (PGCs) and their further role in the pathogenesis of germ cell tumors. Through a review of current literature, based on a comparison of mouse and human models, the development of these cells from an early epiblast to the colonization of the genital ridges is defined. At first text describes the molecular induction of PGCs in response to BMP, WNT, and Nodal signaling pathways, followed by an establishment of germline identity through the tripartite regulatory network of transcription factors: Blimp1, Prdm14, and AP2γ. Special attention is paid to PGCs migration, which involves passive transport resulting from hindgut morphogenesis and active transport regulated by chemotactic signals, particularly the CXCL12/CXCR4 axis and adhesive interactions of the c-Kit/SCF system. Furthermore, the thesis summarizes the process of extensive epigenetic reprogramming specific to PGCs during this stage. The final section focuses on the relationship between migration failure and the formation of ectopic PGCs, which represent a source of extragonadal germ cell tumors.
