Pro analýzu autentického vzorku alkoholického nápoje severokorejského původu (stáří nejméně 12 let) byla použita metoda HPLC založená na gradientové eluci s dvousložkovou mobilní fází, kde složka A byla voda a složka B acetonitril. Separace probíhala na koloně XBridge® BEH C18 (150 × 3,0 mm; 2,5 µm). Metodou HPLC-MS a HPLC-MS/MS bylo v tomto vzorku identifikováno osm ginsenosidů: Rg1, Re, Rf, Rb1, Rc, Rb2, Rd a F2. Tímto krokem proběhla konfirmace toho, že se ve vzorku opravdu nachází kořen z rostliny ženšenu. Metodou RP-HPLC s UV detekcí při 203 nm byly kvantifikovány vybrané ginsenosidy v autentickém vzorku alkoholického nápoje i v referenčním vzorku kořene ženšenu. Dále byly stanoveny vybrané parametry autentického vzorku alkoholického nápoje pro jeho bližší specifikaci. V autentickém vzorku proběhlo stanovení ethanolu metodou GC-FID, stanovení dalších těkavých látek metodou GC×GC-MS, stanovení iontů a sacharidů metodou CZE, stanovení hodnoty pH a stanovení těžkých kovů metodou IPC-MS. Obsah ethanolu v autentickém vzorku byl 26,8 ± 0,6 % obj. Metodou GC×GC/MS byla potvrzena přítomnost těkavých látek typických pro ženšen. For the analysis of an authentic sample of an alcoholic beverage of North Korean origin (at least 12 years old), an HPLC method based on gradient elution with a two-component mobile phase was used, where component A was water and component B was acetonitrile. Separation was performed on an XBridge® BEH C18 column (150 × 3.0 mm; 2.5 µm). Using HPLC-MS and HPLC-MS/MS methods, eight ginsenosides were identified in this sample: Rg1, Re, Rf, Rb1, Rc, Rb2, Rd, and F2. This step confirmed the actual presence of ginseng plant root in the sample. Selected ginsenosides were quantified in both the authentic alcoholic beverage sample and a reference ginseng root sample using the RP-HPLC method with UV detection at 203 nm. Furthermore, selected parameters of the authentic alcoholic beverage sample were determined for its more detailed specification. In the authentic sample, ethanol determination was performed using GC-FID, other volatile substances were determined using GC×GC-MS, ions and carbohydrates were determined using CZE, and the pH value and heavy metals were determined using ICP-MS. The ethanol content in the authentic sample was 26.8 ± 0.6 vol.%. The presence of volatile substances typical for ginseng was confirmed by the GC×GC/MS method.
