[cze][eng] Nikotinové acetylcholinové receptory jsou široce exprimovány v savčím mozku, kde se podílejí na regulaci základních mechanismů, jako je uvolňování neurotransmiterů, neuronová excitabilita anebo synaptická plasticita. Z důvodu svého častého výskytu a funkčnímu významu představují nikotinové receptory atraktivní cíl pro potenciální terapeutické intervence u mnoha neuropsychiatrických onemocnění. Jedním z onemocnění, u kterých se možný terapeutický potenciál nikotinových receptorů aktivně studuje, je také schizofrenie, především její tzv. negativní symptomy. Pacienti trpící schizofrenií mají vyšší riziko silné závislosti na nikotinu a studie na zvířatech prokázaly vztah mezi poruchami nikotinové signalizace a změnami v neuronové aktivitě, které jsou odpovědné za některé behaviorální příznaky relevantní pro lidské pacienty. Cílem této bakalářské práce je popsat podtypy nikotinových receptorů, které se nejčastěji zmiňují v souvislosti s onemocněním schizofrenií a shrnout dostupné poznatky o mechanismech, které podmiňují roli těchto receptorů v patofyziologii tohoto onemocnění. Práce se zaměří nejenom na studie provedené na relevantních zvířecích modelech, ale také na výsledky klinických studií, které byly na toto téma publikovány či jsou registrovány na portále ClinicalTrials.gov. Klíčová slova:...Nicotinic acetylcholine receptors are widely expressed in the mammalian brain, where they help regulate fundamental mechanisms such as neurotransmitter release, neuronal excitability, and synaptic plasticity. Because of their prevalence and functional importance, nicotinic receptors represent an attractive target for potential therapeutic interventions in many neuropsychiatric disorders. One condition in which the possible therapeutic potential of nicotinic receptors is being actively investigated is schizophrenia, particularly its so-called negative symptoms. Patients with schizophrenia are at higher risk of severe nicotine dependence. In addition, animal studies have demonstrated a relationship between disruptions in nicotinic signaling and changes in neuronal activity that are responsible for certain behavioral symptoms relevant to human patients. The aim of this work is to describe the subtypes of nicotinic receptors most frequently mentioned in connection with schizophrenia and to summarize the available findings on the mechanisms underlying the role of these receptors in the pathophysiology of this disorder.
