Zvyšující se počet houbových infekcí způsobených oportunně patogenními kvasinkami rodu Candida v čele s Candida albicans představuje vážný zdravotnický problém. Neúspěšnost léčby kandidóz je podmíněna zejména narůstající rezistencí na běžně dostupná antimykotika, která navíc často vykazují poměrně značnou toxicitu pro lidský organismus. Ze zmíněných důvodů je potřeba hledat nové cíle, na něž by se mohl zaměřit vývoj dalších antimykotik. Jedním z takových cílů by mohla být houbová vakuola a enzymy v ní přítomné. Zatímco vakuolární aspartátová proteáza Pep4p produkovaná Saccharomyces cerevisiae je poměrně dobře prozkoumaná, o homologní proteáze Apr1p kvasinky C. albicans je toho známo velice málo. Cílem této práce bylo prozkoumat a porovnat roli zmíněných proteáz v odpovědi kvasinek na vybrané stresové podmínky a zároveň zjistit jejich vzájemnou zastupitelnost. Jedním ze způsobů, jak toho dosáhnout, bylo testování fenotypu kvasinkových kmenů s deletovanými a zaměněnými geny kódujícími zkoumané proteázy na agarových plotnách, jako druhý způsob byly využity larvy Galleria mellonella, které byly těmito kmeny naočkovány a následně byla sledována jejich mortalita v čase. Dosavadní výsledky této práce naznačují, že vakuolární aspartátová proteáza Apr1p není klíčová pro zvládání vybraných stresových... The increasing incidence of fungal infections caused by opportunistic pathogenic yeasts of the genus Candida, led by Candida albicans, represents a serious healthcare problem. The failure of candidiasis treatment is primarily due to the rising resistance to commonly available antifungal agents, which, moreover, often exhibit significant toxicity to the human organism. For these reasons, it is necessary to identify new drug targets for the development of further antifungals. One such target could be the fungal vacuole and the enzymes present within it. While the vacuolar aspartate protease Pep4p produced by Saccharomyces cerevisiae is relatively well- studied, very little is known about the homologous protease Apr1p of the yeast C. albicans. The aim of this work was to investigate and compare the roles of these proteases in the yeast response to selected stress conditions and to determine their functional substitutability. One approach to achieving this involved phenotypic testing of yeast strains with deleted and switched genes encoding the studied proteases on agar plates. As a second approach, Galleria mellonella larvae were utilized; these were inoculated with the strains, and their mortality was subsequently monitored over time.
