Předložená dizertační práce je zaměřena na přípravu liposomálního komplexu ze stealth liposomů a N,N,N-trimethylchitosanu, který by mohl být využit pro inhalační podávání léčiv s různými fyzikálně-chemickými vlastnosti. Tento systém má poměrně složitou architekturu, a proto byl nejdříve zkoumán vliv jednotlivých složek komplexu i jejich kombinace na změnu fyzikálně-chemických vlastností (velikost, náboj a chování vnitřní a vnější membrány) liposomů. Tvorba liposomálního komplexu byla potvrzena dynamickým a elektroforetickým rozptylem světla, změnou membránových vlastností, metodou kontinuální variace a skenovací transmisní elektronovou mikroskopií. Byl určen i vliv jednotlivých složek liposomálního komplexu na enkapsulační účinnost hydrofobního léčiva erlotinibu, kdy přítomností kyseliny fosfatidové v kompozici liposomů byla dosažena enkapsulační účinnost až 99 %. Nebulizací prázdných liposomů s různým složením i liposomů s enkapsulovaným léčivem různými nebulizačními technikami, charakterizací tohoto aerosolu a depozicí částic v mechanickém i digitálním modelu plic byl vybrán vhodný nebulizátor pro tvorbu aerosolu z liposomálního komplexu. Poslední část dizertační práce byla věnována interakci polymerů, liposomů a celého liposomálního komplexu s plicními surfaktanty. Přítomnost trimethylchitosanu způsobila transformaci struktury plicních surfaktantů z multilamelárních a multivezikulárních systémů na unilamelární. Výsledkem této práce je liposomální komplex schopný enkapsulace hydrofobního léčiva, který je odolný vůči nebulizačnímu kroku se schopností penetrace za sedmé větvení v plicích. Jeho přítomnost v plicních surfaktantech sice způsobí změnu architektury plicních surfaktantů, což nemusí přímo ovlivnit jejich funkčnost.
The presented dissertation is focused on the preparation of a liposomal complex from stealth liposomes and N,N,N-trimethyl chitosan, which could be used for inhalation administration of drugs with different physicochemical properties. This system has a relatively complex architecture, and therefore the effect of the individual components of the complex and their combination on the change in the physicochemical properties (size, charge, and behavior of the inner and outer part of the membrane) of liposomes was investigated. The formation of the liposomal complex was confirmed by dynamic and electrophoretic light scattering, changes in membrane properties, continuous variation method and scanning transmission electron microscopy. The influence of the individual components of the liposomal complex on the encapsulation efficiency of the hydrophobic drug erlotinib was also determined when the encapsulation efficiency of up to 99 % was achieved due to the presence of phosphatidic acid in the composition of liposomes. By nebulizing empty liposomes with different compositions and liposomes with encapsulated drugs using different nebulization techniques, characterizing this aerosol and deposition of particles in a mechanical and digital model of the lungs, a suitable nebulizer was selected to create an aerosol from the liposomal complex. The last part of the dissertation was devoted to the interaction of polymers, liposomes and the entire liposomal complex with lung surfactants. The presence of trimethyl chitosan caused the transformation of the structure of lung surfactants from multilamellar and multivesicular systems to unilamellar. The result of this work is a liposomal complex capable of encapsulating hydrophobic drug that is resistant to the nebulization process with the ability to penetrate beyond the seventh branching in the lungs. Although its presence in lung surfactants caused a change in the architecture of lung surfactants, this may not directly affect their functionality.
