Biosynthetic gene clusters (BGCs) are genomic regions responsible for the production of secondary metabolites with diverse biological activities. Among them, ribosomally synthesized and post-translationally modified peptides (RiPPs) represent a highly diverse class of natural products whose associated gene clusters are often difficult to detect using a single computational approach. This work applies a multi-tool strategy for the detection of RiPP-associated BGCs, evaluating eight bioinformatic tools — including rule-based methods and machine learning–based approaches — on a dataset of fifteen bacterial genomes isolated from Antarctic environments. Prediction outputs were systematically compared and subsequently used for similarity network analysis with BiG-SCAPE in combination with reference clusters from the MIBiG database. Several gene cluster families lacking experimentally characterized reference clusters were identified, suggesting the presence of potentially novel RiPP-associated biosynthetic pathways. The results demonstrate the complementary nature of different prediction approaches and highlight the importance of combining multiple computational tools for a more comprehensive exploration of microbial biosynthetic potential.

což naznačuje přítomnost potenciálně nových RiPP-asociovaných biosyntetických drah. Výsledky ukazují komplementární charakter jednotlivých predikčních přístupů a zdůrazňují význam kombinace více výpočetních metod pro komplexnější průzkum biosyntetického potenciálu mikroorganismů.
