Bakalářská práce se zabývá návrhem a optimalizací planárního biosenzoru s interdigitálními elektrodami (IDE) pro sledování proliferace buněčných kultur a vývojem příslušné měřicí fixtury. První část práce je věnována analýze vlivu geometrie elektrod na citlivost senzoru pomocí simulací v programu Ansys Maxwell. Provedená parametrická analýza prokázala, že s klesající mezerou i šířkou prstů roste intenzita elektrického pole, ale zmenšuje se hloubka jeho průniku. Optimální geometrie by tak měla být volena s ohledem na rozměry analyzovaných buněk. Na základě těchto poznatků byla navržena a realizována deska plošných spojů s měřicí fixturou. Funkčnost navržené desky byla experimentálně ověřena impedančním analyzátorem ve frekvenčním pásmu 20 Hz až 10 MHz. Měření potvrdilo schopnost senzoru spolehlivě rozlišit odlišná média (destilovaná voda a fyziologický roztok) a poukázalo na degradující vliv stejnosměrného předpětí na povrch elektrod ve vodivém prostředí. V závěrečné fázi byla naměřená data aproximována rozšířeným Coleovým modelem, jehož parametry byly extrahovány pomocí optimalizačního algoritmu PSO v prostředí MATLAB, čímž byla potvrzena funkčnost navrženého senzoru.
This bachelor's thesis deals with the design and optimization of a planar biosensor with interdigitated electrodes (IDE) for monitoring cell culture proliferation, and the development of a corresponding measurement fixture. The first part of the thesis is focused on analyzing the influence of electrode geometry on sensor sensitivity using simulations in Ansys Maxwell. The performed parametric analysis proved that with decreasing gap and finger width, the electric field intensity increases, but its penetration depth decreases. Therefore, the optimal geometry should be selected with respect to the dimensions of the analyzed cells. Based on these findings, a printed circuit board with a measurement fixture was designed and realized. The functionality of the proposed system was experimentally verified using an impedance analyzer in the frequency range of 20 Hz to 10 MHz. The measurements confirmed the sensor's ability to reliably distinguish between different media (distilled water and physiological saline) and highlighted the degrading effect of DC bias on the electrode surface in a conductive environment. In the final phase, the measured data were approximated using an extended Cole model, whose parameters were extracted using the PSO optimization algorithm in the MATLAB environment, thereby confirming the functionality of the designed sensor.
