Tato bakalářská práce se zabývá přípravou a charakterizací komplexů agarózové matrice s vybranými vezikulárními systémy. Studovány byly kataniontové IPA vezikuly a lecitinové liposomy, které byly porovnávány jak ve formě čistých vodných disperzí, tak po začlenění do 1 % a 2 % agarózového gelu. Cílem práce bylo posoudit, zda agarózové gelové prostředí ovlivňuje membránové vlastnosti a stabilitu těchto vezikulárních systémů a zda může docházet k narušení nebo rozpadu vezikulární struktury. Čisté disperze byly nejprve charakterizovány pomocí dynamického rozptylu světla a elektroforetického rozptylu světla, které umožnily stanovit velikost částic, polydisperzitu a zeta potenciál. Pro sledování změn membránového mikroprostředí byla použita fluorescenční spektroskopie se sondou Laurdan. Emisní spektra byla měřena v závislosti na teplotě a následně byla vyhodnocena pomocí generalizované polarizace GP. U agarózových vzorků byla spektra korigována s ohledem na optické vlastnosti gelové matrice. Výsledky ukázaly, že agarózová matrice ovlivnila fluorescenční odezvu obou vezikulárních systémů. Vzorky v gelu vykazovaly vyšší hodnoty GP než čisté disperze, což naznačuje méně hydratované a uspořádanější okolí sondy Laurdan.
This bachelor thesis deals with the preparation and characterization of complexes of an agarose matrix with selected vesicular systems. Catanionic IPA vesicles and lecithin liposomes were studied and compared both as pure aqueous dispersions and after incorporation into 1% and 2% agarose gels. The aim of the thesis was to assess whether the agarose gel environment affects the membrane properties and stability of these vesicular systems and whether disruption or disintegration of the vesicular structure may occur. The pure dispersions were first characterized using dynamic light scattering and electrophoretic light scattering, which allowed the determination of particle size, polydispersity, and zeta potential. Fluorescence spectroscopy with the Laurdan probe was used to monitor changes in the membrane microenvironment. Emission spectra were measured as a function of temperature and subsequently evaluated using generalized polarization GP. For agarose samples, the spectra were corrected with respect to the optical properties of the gel matrix. The results showed that the agarose matrix affected the fluorescence response of both vesicular systems. Samples in the gel exhibited higher GP values than pure dispersions, indicating a less hydrated and more ordered environment around the Laurdan probe.
