[cze][eng] Diplomová práce se zabývá vlivem technologie výroby piva na obsah biologicky aktivních látek. Pro účely porovnání byla využita experimentálně vyrobená piva připravená ve výzkumném a výukovém minipivovaru Fakulty zemědělské a technologické Jihočeské univerzity v Českých Budějovicích, a to společně s použitými pivovarskými surovinami. V rámci práce byly analyzovány čtyři druhy světlých piv (11% IPL Kazbek, 11% Kazbek IPA, 11% Pšenica a 12% Pohanka), které se lišily použitými surovinami, technologií kvašení a přidanými složkami. U vzorků byl stanoven obsah vybraných biogenních aminů (tryptamin - TRM, 2-fenylethylamin - PEA, putrescin - PUT, cadaverin - CAD, histamin - HIM, tyramin - TYM, spermidin - SPD a spermin - SPM) a flavonoidu rutinu. TRM ani PEA nebyly detekovány v žádném z analyzovaných piv. Obsahy ostatních sledovaných biogenních aminů (PUT, CAD, HIM, TYM, SPD a SPM) nepřekročily hodnoty uváděné pro srovnatelné produkty v odborné literatuře. Ve všech použitých surovinách byly biogenní aminy prokázány. Pivo 12% Pohanka bylo obohaceno o flavonoid rutin díky přídavku sušené natě pohanky, přičemž jeho koncentrace dosahovala minimálně 28,9 mg/l. Volný kvercetin nebyl v tomto pivu detekován v průběhu zrání ani skladování. Získané výsledky naznačují, že technologie minipivovarů nepodporuje zvýšenou tvorbu biogenních aminů v pivu. Současně potvrzují možnost výroby piva obohaceného o biologicky aktivní látky, například flavonoidy, v podmínkách minipivovarské produkce. Společně s chemickou analýzou vybraných biogenních aminů byl stanoven senzorický profil piv 11% IPL Kazbek, 11% Kazbek IPA a 11% Pohanka za účelem posouzení, zda technologie výroby ovlivňuje senzorické vlastnosti. Hodnoceny byly vybrané senzorické charakteristiky zahrnující vzhled, vůni a chuť piva. Mezi analyzovanými vzorky byly zjištěny jak výraznější, tak minimální rozdíly. Všechna hodnocená piva byla posouzena jako chuťově přijatelná s pozitivním celkovým dojmem. Nejvýraznější rozdíly byly zaznamenány v charakteru hořkosti, zatímco nejmenší v barvě piva. Výsledky naznačují, že technologie výroby se v senzorickém profilu piv projevuje, avšak její vliv je spíše omezený. This thesis examines the influence of beer production technology on the content of biologically active substances. For comparison purposes, experimentally brewed beers prepared at the research and teaching microbrewery of the Faculty of Agriculture and Technology at the University of South Bohemia in České Budějovice were used, along with the brewing ingredients employed. The study analyzed four types of light beers (11% IPL Kazbek, 11% Kazbek IPA, 11% Pšenica, and 12% Pohanka), which differed in the raw materials used, fermentation technology, and added ingredients. The content of selected biogenic amines (tryptamine - TRM, 2-phenylethylamine - PEA, putrescine - PUT, cadaverine - CAD, histamine - HIM, tyramine - TYM, spermidine - SPD, and spermine - SPM) and the flavonoid rutin was determined in the samples. Neither TRM nor PEA were detected in any of the analyzed beers. The contents of the other monitored biogenic amines (PUT, CAD, HIM, TYM, SPD, and SPM) did not exceed the values reported for comparable products in the scientific literature. Biogenic amines were detected in all raw materials used. The 12% Buckwheat beer was enriched with the flavonoid rutin through the addition of dried buckwheat leaves, with its concentration reaching at least 28.9 mg/l. Free quercetin was not detected in this beer during maturation or storage. The results obtained suggest that microbrewery technology does not promote increased formation of biogenic amines in beer. At the same time, they confirm the possibility of producing beer enriched with biologically active substances, such as flavonoids, under microbrewery production conditions. Along with the chemical analysis of selected biogenic amines, the sensory profile of the 11% IPL Kazbek, 11% Kazbek IPA, and 11% Pohanka beers was determined to assess whether the production technology affects sensory properties. Selected sensory characteristics, including the beer's appearance, aroma, and taste, were evaluated. Both significant and minimal differences were found among the analyzed samples. All evaluated beers were assessed as palatable with a positive overall impression. The most pronounced differences were observed in bitterness, while the smallest differences were found in beer color. The results suggest that production technology does influence the sensory profile of the beers, but its effect is rather limited.
