Tato bakalářská práce se zabývá izolací a následnou frakcionací arteficiálních huminových látek připravených z odpadní biomasy, přesněji kávové sedliny. Tyto látky byly následně podrobeny několika vybraným metodám určených ke kvantifikaci a fyzikálně-chemické charakterizaci arteficiálních huminových látek. Vzorky byly analyzovány spektrometrickými metodami, konkrétně FTIR, UV/Vis a fluorescenční spektrometrií a termickými metodami, tedy elementární a termogravimetrickou analýzou. Vyhodnocení těchto metod slouží k získání cenných informací o kvalitě frakcionovaných arteficiálních huminových látek, o struktuře a fyzikálně-chemických vlastnostech těchto látek. Z výsledků je patrné, že připravené arteficiální huminové látky z kávové sedliny jsou vlastnostmi a povahou srovnatelné s huminovými látkami z přírodních matric, jako jsou například kaustobiolity. Nicméně byl pro látky zaznamenán nízký stupeň humifikace. Na základě získaných výsledků s ohledem na množství popela byla navržena optimalizace izolačních, frakcionačních a purifikačních postupů ke zvýšení kvality izolovaných vzorků.
This bachelor’s thesis focuses on the isolation and subsequent fractionation of artificial humic substances prepared from waste biomass, specifically coffee grounds. These substances were then subjected to several selected methods designed for the quantification and physicochemical characterization of artificial humic substances. The samples were analyzed using spectroscopic methods, specifically FTIR, UV/Vis, and fluorescence spectroscopy, as well as thermal methods, namely elemental and thermogravimetric analysis. The evaluation of these methods serves to obtain valuable information about the quality of the fractionated artificial humic substances, as well as their structure and physicochemical properties. The results show that the prepared artificial humic substances derived from coffee grounds are comparable in properties and nature to humic substances from natural matrices, such as kaustobiolites. However, a low degree of humification was observed for these substances. Based on the results obtained, with regard to ash content, an optimization of the isolation, fractionation, and purification procedures was proposed to improve the quality of the isolated samples.
