Toxocara canis je střevní parazit psovitých šelem rozšířený po celém světě. U člověka může způsobit vážné klinické projevy známé jako larvální toxokaróza, které se v případě výskytu v centrální nervové soustavě označují jako cerebrální toxokaróza. Experimentální výzkumy na myších pomáhají osvětlit problematiku této infekce. Tato práce je zaměřena na vyhodnocení vlivu infekce myší různými infekčními dávkami larev T. canis na jejich protilátkovou odpověď, fyzický stav a motorické schopnosti v kombinaci s patogenezí v jejich mozcích. Další otázkou je, zda nízká infekční dávka odpovídající infekcím v přírodě způsobuje u infikovaných myší shodné změny jako vysoká infekční dávka, pokud je působení larev T. canis na hostitele dostatečně dlouhé. Myši BALB/c byly ve dvou experimentech orálně infikovány 10, 100, případně 1000 larvami T. canis a pravidelně pozorovány a testovány do 97. a 100. týdne po infekci. V obou experimentech byl hodnocen celkový vzhled, motorické schopnosti a jejich změny detekované jak pouhým pozorováním, tak i sérií testů. Po odběru materiálu byly na histologických řezech zhodnoceny patologické změny mozkové tkáně. Výsledkem bylo zjištění mírných až závažných poruch celkového stavu a motorických schopností přímo úměrných velikosti infekční dávky a rychlosti rozvoje těchto poruch u... Toxocara canis is an intestinal parasite of canids that is widespread throughout the world. In humans, it can cause serious clinical symptoms known as larval toxocariasis, which, when occurring in the central nervous system, is referred to as cerebral toxocariasis. Experimental research on mice helps shed light on the problem of this infection. This work focuses on evaluating the effect of infecting mice with different doses of T. canis larvae on their antibody response, physical condition, and motor skills in combination with pathogenesis in their brains. Another question is whether a low dose corresponding to infections in nature causes the same changes in infected mice as a high infectious dose, if the effect of T. canis larvae on the host is sufficiently long. BALB/c mice were orally infected with 10, 100, or 1000 T. canis larvae in two experiments and regularly observed and tested until 97 and 100 weeks after infection. In both experiments, overall appearance, motor skills, and changes in these were evaluated both by simple observation and a series of tests. After collection of the material, pathological changes in brain tissue were evaluated on histological sections.
