Tato bakalářská práce se zabývá navržením metodiky stanovení amoniakálního dusíku v reálných matricích, zejména odpadních produktech bioplynové stanice. Byl vybrán nejvhodnější postup stanovení s ohledem na matrici vzorku, laboratorní instrumentaci, rychlost, citlivost a náročnost. Navržená metodika se skládá ze dvou po sobě jdoucích kroků, kterými je destilace a následné spektrofotometrické stanovení pomocí mobilních souprav. Destilace je výbornou metodou pro separaci amoniakálního dusíku díky její selektivitě. Využitím spektrofotometrické metody se docílí, v porovnání s volumetrickou metodou, snížení mezi stanovitelnosti, a použitím mobilních souprav, které jsou svými výsledky srovnatelné s laboratorním stanovením, se zkrátí doba analýzy. Metodika byla optimalizována pro dosažení přesných a správných výsledků. K ověření metodiky byly analyzovány vzorky digestátu, který je odpadním produktem bioplynové stanice. Je to nehomogenní materiál, bohatý na amoniakální dusík. Pomocí optimalizované metodiky bylo stanoveno procentuální hmotnostní zastoupení amoniakálního dusíku v těchto vzorcích, které se pohybovalo v rozmezí 0,18-0,55 %. Se snižující se homogenitou digestátu klesalo procentuální zastoupení amoniakálního dusíku. This bachelor thesis focuses on the design of a methodology for the determination of ammoniacal nitrogen in real matrices, especially biogas plant waste products. The most appropriate determination procedure was selected with regard to sample matrix, laboratory instrumentation, speed, sensitivity and difficulty. The suggested methodology consists of two consecutive steps, which are distillation and subsequent spectrophotometric determination using mobile kits. Distillation is an excellent method for the separation of ammoniacal nitrogen due to its selectivity. The use of the spectrophotometric method results in a reduction of the limit of quantification compared to the volumetric method, and the use of mobile kits, which are comparable to the laboratory determination, reduces the analysis time. The methodology has been optimised to achieve accurate and correct results. To verify the methodology, samples of digestate, which is the waste product of a biogas plant, were analysed. It is an inhomogeneous material, rich in ammoniacal nitrogen. Using the optimised methodology, the percentage by weight of ammonia nitrogen in these samples was determined and ranged from 0.18-0.55 %. The percentage of ammonia nitrogen decreased with decreasing digestate homogeneity.
