Schisandra chinensis je divoce rostoucí liána pocházející z východní Asie. Je zařazena mezi rostlinné adaptogeny a její plody jsou v současnosti používány pro produkci potravinových doplňků s pozitivními účinky na lidské zdraví díky obsahu různých sekundárních metabolitů. Lignan schisandrin je dominantní bioaktivní látkou v plodech. Na druhou stranu lignany jsou známé pro své alelopatické nebo toxické účinky na jiné organismy (rostliny, zvířata). S. chinensis není v evropských ekosystémech přirozená a její ekotoxikologické vlastnosti, zejména v případě její intenzivní zemědělské produkce, nebyly dosud hodnoceny. Pomocí Soxhletovy extrakce byl připraven vodný extrakt z plodů S. chinensis, který simuloval přirozené vylouhování aktivních látek ze zralých plodů spadených do vody nebo na půdu. Extrakt byl použit v koncentracích 0.045 až 45 mg/L podle obsahu schisandrinu. Pro testy ekotoxicity byly vybrány modelové organismy Lemna minor, Daphnia magna a Thamnocephalus platyurus reprezentující vodní prostředí a kulturní plodiny Panicum miliaceum (jednoděložné) a Sinapis alba (dvouděložné) jako zástupci suchozemského prostředí. Během sedmidenní kultivace nízké koncentrace (0.045 a 0.09 mg/L) extraktu z S. chinensis nezpůsobily žádné změny v růstových parametrech nebo fotosyntetickém výkonu L. minor. Vyšší zatížení (0.45 a 0.9 mg/L) vyvolala signifikantní omezení v počtu rostlin, celkové listové ploše a čerstvé hmotnosti. Fotosyntetické parametry (základní fluorescence chlorofylu, kvantové výtěžky) byly ovlivněny pouze koncentrací 0.9 mg/L. Nejvyšší koncentrace 45 mg/L byla extrémně toxická pro rostliny okřehku a způsobila jejich smrt během prvních pěti dnů kultivace. Efektivní koncentrace (EC50) způsobující 50% letální účinek na D. magna byla stanovena na 0.045 mg/L po 24 hodinách a 0.015 mg/L po 48 hodinách. EC50 pro T. platyurus dosáhla 0.46 mg/L po 24 hodinách, tedy více než desetinásobně vyšší hodnota oproti D. magna. V případě suchozemských rostlin byla fytotoxicita extraktu testována na klíčivosti semen a růstu klíčních rostlin P. miliaceum a S. alba. Pouze nejvyšší koncentrace schisandrinu způsobila inhibici klíčení semen a růstu kořene a nadzemní části u obou druhů. Účinek nízkých koncentrací byl zanedbatelný a přídavek živin nepřispěl významně k potlačení negativního vlivu schisandrinu. Tato práce ukázala, že potenciální environmentálně relevantní koncentrace bioaktivních látek z S. chinensis může představovat vážné riziko pro vodní i suchozemské ekosystémy.
Schisandra chinensis is a wild east-Asian liana plant. It is classified as a plant adaptogen and its fruits are currently used to produce nutrient supplements with positive effects on human health due to the content of various secondary metabolites. Lignan schisandrin is the dominant bioactive compound in fruits. On the other hand, lignans are known for their possible allelopathic or toxic effects concerning other organisms (plants, animals). S. chinensis is not native to European ecosystems, and its ecotoxicological properties, especially in the case of its intensive agricultural production, have not been verified yet. Using Soxhlet extraction the crude water extract from S. chinensis fruits was prepared to simulate the natural leakage of active substances from ripe fruits fallen into the water or soil. The extract was used in treatments from 0.045 to 45 mg/L according to the content of schisadrin. For ecotoxicity tests, Lemna minor, Daphnia magna, and Thamnocephalus platyurus were selected as suitable model organisms representing the aquatic environment and crops Panicum miliaceum (monocot) and Sinapis alba (dicot) as representatives from the terrestrial environment. During seven days of cultivation, in low treatments (0.045 and 0.09 mg/L), the extract of S. chinensis did not cause any changes in L. minor growth parameters or photosynthetic performance. Higher treatments (0.45 and 0.9 mg/L) caused significant limitations in plants’ number, total leaf area, and fresh weight. The photosynthetic parameters (basal chlorophyll fluorescence, quantum yields) were affected only by 0.9 mg/L. The highest treatment, 45 mg/L, exhibited extreme toxicity to duckweed plants causing their death during the first five days of cultivation. Effective concentration (EC50) causing 50% lethal effect for D. magna was established to 0.045 mg/L after 24 h and 0.015 mg/L after 48 h. EC50 for T. platyurus reached 0.46 mg/L after 24 h, i.e. more than ten times higher than for D. magna. In the case of terrestrial plants the extract´s phytotoxicity was tested based on seed germination and seedling growth of P. miliaceum and S. alba. Only the highest concentration of schisandrin caused inhibition of seed germination and root and shoot growth in both species. The effect of lower concentrations was negligible, and the addition of nutrients did not contribute considerably to suppressing the negative influence of schisandrin. This work showed that the potential environmentally relevant concentrations of S. chinensis bioactive compounds could represent a severe risk for both aquatic and terrestrial ecosystems.
