Cévní mozková příhoda (CMP) je závažný zdravotní stav, který může pacienta ohrozit na životě nebo mu způsobit trvalé následky. Se stále se zvyšující incidencí tohoto onemocnění stoupá i potřeba používání adekvátních a bezpečných trombolytik pro terapii. Tato práce se zaměřuje na modifikaci struktury a potenciální zlepšení vlastností v současné době nejpoužívanějšího trombolytika - alteplasy. Cílem práce je příprava konjugátu alteplasa-polymer tak, aby došlo k prodloužení účinku samotného trombolytika. Příprava konjugátu navazuje na zkušenosti kolegů z oddělení Biolékařských polymerů ÚMCH AV ČR, kteří pracovali na modifikaci různých funkčních skupin alteplasy. Tato práce se zaměřuje na modifikaci alteplasy novou cestou, kdy dochází k oxidaci manosových jednotek proteinu a následnému navázání alteplasy na vhodný polymerní nosič. Konjugací s polymerem by mělo dojít k prodloužení cirkulace trombolytika v krevním řečišti, a tím i jeho biologického účinku. Jedním z mechanismů by mohlo být i zpomalení biologického odbourávání alteplasy v játrech v důsledku oxidace manosových jednotek, které bez modifikace běžně reagují s manosovými receptory hepatocytů. V rámci diplomové práce byly připraveny polymerní nosiče, přečištěním lékové formy byla získána čistá alteplasa a následně byly syntetizovány polymerní... Stroke is a serious medical condition that can be life-threatening or cause permanent damage. With the incidence of this disease continuing to rise, there is an increasing need for adequate and safe thrombolytic agents for treatment. Thisworkfocusesonmodifyingthestructureandpotentiallyimprovingthepropertiesofthemost commonly used thrombolytic agent today - alteplase. The aim of the work is to prepare an alteplase- polymer conjugate in order to prolong the effect of the thrombolytic agent itself. The preparation of the conjugate builds on the experience of colleagues from the Department of Biomedical Polymers at the Institute of Macromolecular Chemistry of the Czech Academy of Sciences, who worked on the modification of various functional groups of alteplase. This work focuses on the modification of alteplase in a new way, involving the oxidation of mannose units of the protein and the subsequent binding of alteplase to a suitable polymer carrier. Conjugation withthe polymer shouldprolongthe circulationof the thrombolytic agent in the bloodstream and thus its biological effect. One of the mechanisms could be the slowing downofthe biological degradationof alteplasein theliveras a result ofthe oxidation ofmannose units, which normally react with mannose receptors in hepatocytes without modification....
