[cze][eng] Úvod: Obezita představuje zásadní problém veřejného zdraví s narůstající prevalencí u dospělých i dětí. Její etiopatogeneze je multifaktoriální a zahrnuje interakci genetických, environmentálních a socioekonomických faktorů ovlivňujících ukládání tělesného tuku i odolnost těchto zásob vůči redukčním intervencím. Genetická predispozice se na regulaci tělesné hmotnosti podílí zhruba z 40-70 %. Výzkum genetického pozadí obezity, zejména prostřednictvím celogenomových asociačních studií (GWAS), doposud identifikoval více než tisíc lokusů spojených s obezitou, z nichž mnohé byly potvrzeny i u dětské populace. Rostoucí prevalence dětské obezity však ukazuje, že samotná znalost genetických faktorů nebude ke zvládání již existujícího problému stačit. Stále větší pozornost se proto soustředí na zkoumání vlivu genetických predispozic na úspěšnost redukčních intervencí, což může přispět k rozvoji účinnějších terapeutických strategií. Tato diplomová práce navazuje na předchozí výzkum a rozšiřuje dosavadní poznatky analýzou vlivu vybraných genetických variant na úspěšnost krátkodobé intervence životního stylu u obézních dětských pacientů. Cíl: Hlavním cílem této diplomové práce je analýza vlivu genetických variant SLC39A8 (rs13107325), SEC16B (rs543874) a GNPDA2 (rs10938397) na úspěšnost lázeňské léčebné...Introduction: Obesity represents a major public health issue with a rising prevalence among both adults and children. Its etiopathogenesis is multifactorial, involving interactions of genetic, environmental, and socioeconomic factors that influence body fat accumulation and the resistance of fat stores to reduction interventions. Genetic predisposition accounts for approximately 40-70% of body weight regulation. Research into the genetic background of obesity, particularly through genome-wide association studies (GWAS), has identified over a thousand loci associated with obesity, many confirmed also in children. The increasing prevalence of childhood obesity indicates that knowledge of genetic factors alone is insufficient to address the existing problem. Growing attention is therefore directed toward examining how genetic predispositions affect the success of weight-reduction interventions, which may support the development of more effective therapeutic strategies. This thesis builds on previous research and expands current knowledge by analyzing the influence of selected genetic variants on the effectiveness of a short-term lifestyle intervention in obese pediatric patients.
