Cílem této diplomové práce bylo ověřit účinnost hygienizace kalu ze septiku kořenové čistírny odpadních vod o velikosti 150 ekvivalentních obyvatel pomocí kombinace mléčné fermentace a následného vermikompostování, tedy tzv. Terra Preta sanitace. Porovnávány byly tři varianty uspořádání - KAL (kontrola), KV (kal smíchaný s odpadními výlisky z přípravy čerstvých džusů), KVP (kal s výlisky z přípravy džusů a pilinami). Před fází vermikompostování byla každá varianta ještě rozdělena na dvě podvarianty - bez přídavku a s přídavkem biouhlu. Účinnost zvolené metody byla hodnocena na základě změny v koncentraci indikátorového mikroorganismu Escherichia coli, monitorována byla však také koncentrace koliformních a termotolerantních koliformních bakterií, intestinálních enterokoků a bakterií mléčného kvašení. Současně byl v jednotlivých fázích procesu monitorován obsah vybraných mikropolutantů ze skupiny PPCPs a hodnota pH. U fáze vermikompostování byly navíc sledovány parametry jako vitalita žížal a počet kokonů. Mikrobiologické analýzy byly provedeny pomocí kultivačních metod za použití selektivních půd. Ke konfirmaci izolovaných kolonií, které byly vyhodnoceny jako enterokoky, byla využita metoda MALDI TOF. Koncentrace mikropolutantů ze skupiny PPCPs byla po extrakci vzorků změřena s využitím LC/MS... The aim of this thesis was to verify the effetiveness of sanitation of septic tank sludge from constructed wetland wastewater treatment plant designed for 150 population equivalents, using a combination of lactic acid fermentation and subsequest vermicomposting, i.e., the so- called Terra Preta sanitation. Three arrangement variants were compared: KAL (control), KV (sludge mixed with waste pulp from fresh juice preparation), and KVP (sludge with juice pulp and sawdust). Before the vermicomposting phase, each variant was divided into two sub-variants - without the addition of biochar and with the addition of biochar. The effectiveness of the chosen method was evaluated based on changes in the content of the indicator microorganism Escherichia coli. Additionally, coliform and thermotolerant coliform bacteria, intestinal enterococci, and lactic acid bacteria were monitored. The content of selected micropollutants from the PPCPs group and pH values were also monitored throughout the individual phase of the process. In the vermicomposting stage, further parameters such as earthworm vitality and cocoon count were assessed. Microbiological analyses were performed using culture-based methods with selective media.
