[cze][eng] Flaviviry představují významnou skupinu RNA virů zahrnující řadu lidských patogenů, mezi něž patří mimo jiné i virus dengue. Infekce virem dengue každoročně postihuje desítky milionů lidí po celém světě a v současnosti pro ni neexistuje specifická antivirová terapie. Replikace virového genomu je zajišťována RNA-dependentní RNApolymerázou (RdRp), která tvoří C-terminální doménu multifunkčního proteinu NS5 a představuje atraktivní cíl pro vývoj nových antivirových léčiv. Tato diplomová práce se zaměřila na testování synteticky upravených nukleotidů jako potenciálních inhibitorů polymerázové aktivity RdRp domény proteinu NS5 z viru dengue typu 2. RdRp byla exprimována v bakteriálním systému E. coli a purifikována kombinací afinitní, iontově-výměnné a gelové chromatografie. Její aktivita byla analyzována pomocí in vitro polymerázových esejí s RNA substrátem, přičemž produkty reakce byly analyzovány denaturační polyakrylamidovou gelovou elektroforézou. Inhibiční účinek testovaných látek byl hodnocen v kompetitivních a nekompetitivních inhibičních esejích, které umožnily také získat základní informace o možném mechanismu jejich účinku ve vztahu k přirozeným nukleosidtrifosfátům. Z výsledků vyplynulo, že část testovaných látek vykazuje inhibiční účinek vůči RdRp doméně proteinu NS5, přičemž u vybraných...Flaviviruses represent an important group of RNA viruses that include several human pathogens, among which dengue virus is one of the most significant. Dengue virus infection affects tens of millions of people worldwide each year, and currently no specific antiviral therapy is available. Replication of the viral genome is carried out by the RNA-dependent RNA polymerase (RdRp), which forms the C-terminal domain of the multifunctional NS5 protein and represents an attractive target for the development of new antiviral agents. This thesis focused on testing synthetically modified nucleotides as potential inhibitors of the polymerase activity of the RdRp domain of the NS5 protein from dengue virus type 2. The RdRp was expressed in an E. coli system and purified using a combination of affinity, ion- exchange, and gel chromatography. Its activity was analyzed using in vitro polymerase assays with an RNA substrate, and the reaction products were analyzed by denaturing polyacrylamide gel electrophoresis. The inhibitory effects of the tested compounds were evaluated in competitive and non-competitive inhibition assays, which also provided basic information on the possible mechanism of their action in relation to natural nucleoside triphosphates.
