Cílem této bakalářské práce bylo zjistit, zda existuje souvislost mezi výskytem polymorfismů C677T a A1298C v genu MTHFR a A66G v genu MTRR a výskytem idiopatické neplodnosti. Téma bylo zvoleno s ohledem na aktuální výzkumy, které poukazují na možnou souvislost poruch metabolismu folátů a zvýšené hladiny homocysteinu s reprodukčními poruchami. Teoretická část se zaměřuje na přehled příčin neplodnosti, význam metabolismu folátu a homocysteinu, úlohu enzymů MTHFR a MTRR a vliv jejich genových variant na enzymatickou aktivitu a hladinu homocysteinu v organismu. Dále jsou podrobně popsány principy molekulárně-biologických metod, které byly využity při analýze genetického materiálu. Praktická část byla realizována v genetické laboratoři GENLABS a zahrnovala zpracování celkem 444 biologických vzorků (bukální stěr a periferní krev). DNA byla izolována a následně analyzována pomocí PCR, ARMS-PCR, real-time PCR a gelové elektroforézy. U všech vzorků byla provedena genotypizace genu MTHFR, u části vzorků také analýza genu MTRR. Výsledky ukázaly častý výskyt sledovaných polymorfismů a některé kombinace variant, které mohou ovlivňovat hladiny homocysteinu a tím potenciálně přispívat k rozvoji idiopatické neplodnosti. Přínosem práce je nejen zpracování rozsáhlého souboru dat, ale také potvrzení významu genetických faktorů při hodnocení fertility. Výsledky podporují využití genotypizace jako doplňkového nástroje v diagnostice neplodnosti a zdůrazňují potenciál personalizovaného přístupu ke klinické léčbě v rámci reprodukční medicíny. The aim of this bachelor thesis was to determine whether there is an association between the occurrence of the C677T and A1298C polymorphisms in the MTHFR gene and the A66G polymorphism in the MTRR gene with idiopathic infertility. The topic was chosen in the light of recent research that suggest a potential link between disturbances in folate metabolism and elevated homocysteine levels with reproductive disorders. The theoretical part focuses on an overview of the causes of infertility, the importance of folate and homocysteine metabolism, the role of the MTHFR and MTRR enzymes, and the impact of their genetic variants on enzymatic activity and homocysteine levels in the body. In addition, the principles of molecular biological methods used for genetic analysis are described in detail. The practical part was carried out in the GENLABS genetic laboratory and included the processing of a total of 444 biological samples (buccal swabs and peripheral blood). DNA was isolated and subsequently analyzed using PCR, ARMS-PCR, real-time PCR, and gel electrophoresis. Genotyping of the MTHFR gene was performed on all samples, and MTRR gene analysis was conducted on a subset of samples. The results showed a frequent occurrence of the studied polymorphisms and some variant combinations that may influence homocysteine levels, thus potentially contributing to the development of idiopathic infertility. The contribution of this thesis lies not only in the analysis of a large dataset but also in confirming the significance of genetic factors in the assessment of fertility. The results support the potential use of genotyping as a supplementary tool in the diagnosis of infertility and highlight the relevance of personalized approaches in clinical reproductive medicine.
