Abstract: This thesis explores the role of ribokinase (RBKS) in the immunometabolism of Drosophila melanogaster, with particular focus on its potential involvement in the newly proposed gluconate ribose pathway (GRP). Using genetic tools, RBKS mutants were generated and tested for immune and metabolic phenotypes. Preliminary immune assays, including lamellocyte counts and resistance to parasitoid wasp infection, suggest a role of RBKS in immunity. Mass spectrometry revealed metabolic changes, including altered nucleotide synthesis and accumulation of 2 dehydrogluconate, a key GRP intermediate. These findings support a metabolic function of RBKS and highlight the GRP as a potential link between carbohydrate metabolism and immune function in Drosophila.
