[cze][eng] Tato bakalářská práce se zabývá genetickou detekcí trombofilních stavů. Zabývá se především Leidenskou mutací a mutací v genu pro protrombin. Tyto dvě mutace jsou zároveň nejčetněji zastoupené v naší populaci. V teoretické části této práce jsou vysvětleny pojmy hemostáza, trombofilní stavy, Leidenská mutace a mutace v genu pro protrombin. Trombofilní stavy patří mezi rutinně vyšetřované a mezi nejčastější patří Leidenská mutace. Leidenská mutace se řadí mezi vrozené trombofilie a vyskytuje u mužů i u žen. Větší riziko trombofilního stavu ale nesou více ženy než muži. Leidenská mutace je mutace genu pro srážlivý faktor V, který způsobuje velké riziko krevních sraženin. Mutace v genu pro protrombin je mutace srážlivého faktoru II, který způsobuje taktéž velké riziko krevních sraženin. Praktická část této bakalářské práce je zaměřena na analýzu Leidenské mutace a mutace v genu pro protrombin u probandů. Celkem bylo pro tuto práci odebráno 62 vzorků nesrážlivé periferní krve. DNA byla vyizolována z periferní krve, následně byla změřena její koncentrace ve spektrofotometru a Leidenská mutace (1691G> A) i mutace pro protrombin (20210G> A) byla detekována prostřednictvím Q-PCR.This bachelor's thesis focuses on the genetic detection of thrombophilic conditions, primarily addressing the Leiden mutation and mutations in the prothrombin gene. These two mutations are also the most common in our population. The theoretical part of this thesis explains concepts such as hemostasis, thrombophilic conditions, the Leiden mutation, and mutations in the prothrombin gene. Thrombophilic conditions are among those routinely investigated, with the Leiden mutation being one of the most frequent. The Leiden mutation is classified as an inherited thrombophilia and occurs in both men and women. However, women are at a greater risk of thrombophilic conditions than men. The Leiden mutation is a mutation of the gene for coagulation factor V, which poses a significant risk of blood clots. Mutations in the prothrombin gene involve a mutation of coagulation factor II, also posing a significant risk of blood clots. The practical part of this bachelor's thesis focuses on the analysis of the Leiden mutation and mutations in the prothrombin gene in probands. A total of 62 samples of non-coagulated peripheral blood were collected for this thesis. DNA was isolated from peripheral blood, its concentration was measured using a spectrophotometer, and the Leiden mutation (1691G> A) and prothrombin mutation (20210G> A) were detected using Q-PCR.
