Abstract: The project investigates the mechanistic advantage of the evolved front proton entry site blockage in mitochondrial ATP synthases of the flagellate protist Trypanosoma brucei. To study the effects of unblocking in both directions of ATP synthase activity, the subunit-aN of procyclic and bloodstream forms were modified using current gene editing tools. The stability of native mitochondrial protein complexes with respect to this genetic engineering was then assessed, highlighting the ways in which further experimentation with such essential genes can be enhanced.
