Abstract: The research goal is to identify cardiolipin remodeling enzyme. For the purpose of my study, we chose two proteins: ALCAT1 and delta 12 desaturase. ALCAT1 will be epitope-tagged to determine its subcellular localization in T. brucei. Its expression will be depleted by RNA interference to see if there is any effect on the physiology and morphology of the mitochondrion. Delta 12 desaturase may be involved in the remodeling of all phospholipids not just cardiolipins. To see if there is any effect on the stage differentiation and growth we will deplete the expression of delta 12 desaturase by gene knockout.
