Hlavním cílem této práce je vytipování molekulárních markerů pro určení genetické variability u pošvatky Diura bicaudata z Moravskoslezských Beskyd. DNA byla úspěšně naamplifikována pomocí mitochondriálních markerů COI a 16S. Získané sekvence byly použity pro populačně genetické analýzy. Nebyla objevena žádná struktura populací a bylo prokázáno, že migrace převažuje genetickým driftem. Bylo taktéž potvruzeno, že všechny zkoumané vzorky jsou Diura bicaudata. The main aim of this thesis was to identify molecular markers that will help to determine the genetic variability among populations of stonefly Diura bicaudata from the Moravskozlezské Beskydy. DNA was successfully amplified using mitochondrial markers COI and 16S. The obtained sequences were used for genetic analysis of populations. No population structure has been demonstrated and migration over genetic drift has been confirmed. It was confirmed that the examined specimens belong to Diura bicaudata.
