[eng][cze] Wetlands, wet meadows and calcareous fens are currently considered as some of the most endangered habitats in Europe. The number and area of these habitats have decreased due to land use changes (intensification, drainage, abandonment of traditional management such as mowing). The same fate has befallen species associated with these habitats. This work deals with the population biology and genetics of two species associated with these habitats. The first species (Tofieldia calyculata) is a critically endangered species, whose lowland populations are almost extinct, although it is relatively abundant in mountain habitats of Central Europe. The aim of this PhD thesis was to determine whether lowland populations are genetically different from mountain populations and what genetic diversity remains in lowland populations. The second aim focused on differences in the population biology of the species in different habitat types (fen meadows compared to mountain habitats). Mountain populations have higher genetic diversity than lowland populations. It was not possible to clearly confirm the difference of lowland populations. On the other hand, some parameters of genetic diversity indicate the possible relict status and difference of these populations, although some lowland populations arose as a result of...Mokřady, vlhké a slatinné louky jsou v současnosti považovány za jedny z nejohroženějších biotopů v Evropě. V důsledku změn v zemědělském hospodaření (intenzifikace zemědělství, odvodňování nebo naopak upuštění od tradiční péče jako bylo kosení) jejich počet výrazně poklesl a stejný osud potkal druhy na tyto biotopy vázané. Tato práce se zabývá populační biologií a genetikou dvou druhů vázaných na tyto biotopy. Kohátka kalíškatá (Tofieldia calyculata) je kriticky ohrožený druh, jehož nížinné populace jsou na pokraji vyhynutí, přestože v okolních zemích je v horských biotopech poměrně hojná. Cílem dizertační práce bylo zjistit, zda jsou nížinné populace geneticky odlišné od horských a jaká v nich zbyla genetická diverzita. Druhý neméně podstatný cíl se zaměřil na rozdíly v populační biologii druhu v různých prostředích (slatinné louky ve srovnání s horskými biotopy). Horské populace mají vyšší genetickou diverzitu než populace nížinné. Nepodařilo se jednoznačně potvrdit odlišnost nížinných populací. Na druhou stranu některé parametry genetické diverzity naznačují možnou reliktnost a odlišnost těchto populací, byť část nížinných populací vznikla jako důsledek rozšíření po skončení poslední doby ledové.
