[cze][eng] Souhrn Disertační práce se zaměřuje na problematiku diagnostiky kožních nádorů, která se v dnešní době opírá o chirurgickou excizi a histopatologickou analýzu. Jedná se již o invazivní vyšetření, jenž sebou nese i jistá rizika a je spojeno s několika úskalími z pohledu pacienta, tak i zdravotního systému. Snahou této práce byla implementace spektroskopie laserem buzeného plazmatu (LIBS) a hmotnostní spektrometrie indukčně vázaného plazmatu s laserovou ablací (LA-ICP-MS) v diagnostice kožních nádorů. Cílem výzkumu bylo ověřit možnost odlišit maligní tkáň ve vzorku na základě odlišného chemického složení pomocí zmíněných dvou analytických technik. Práce se soustředila na optimalizaci metodiky, přípravy a zpracování vzorků, optimalizace metody laserové ablace a optimalizaci metodiky pro zpracování dat a jejich interpretaci. Součástí bylo korelativní zobrazování hyperspektrálních dat spektroskopie a spektrometrie s histologickou analýzou a její komplexnější provázání s biologií tkáně. Získané výsledky vybraných biogenních prvků (např. Ca, Mg, Zn, P) poukazují na potenciál laserové spektroskopie a spektrometrie identifikovat nádorovou tkáň. Práce přináší významný posun v interpretaci dat a jejich komplexnějšího zhodnocení napříč různými tumory i daného jednoho tumoru. Studie přinesla významný interdisciplinární...The dissertation thesis focuses on the issue of skin cancer diagnosis, which nowadays relies on surgical excision and histopathological analysis. This is an invasive examination that carries certain risks and presents several challenges from both the patient's and the healthcare system's perspectives. This work aimed to implement laser-induced breakdown spectroscopy (LIBS) and laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) in the diagnosis of cutaneous malignancies. The goal of this research was to verify the possibility of distinguishing malignant tissue in a sample based on its different chemical composition using the two analytical techniques mentioned above. The research explored optimizing the methodology, preparation, and processing of samples, laser ablation method optimization, and refining data processing and interpretation. It included correlative hyperspectral imaging data from spectroscopy and spectrometry with histological analysis and a more complex link with tissue biology. The obtained results of selected biogenic elements (e.g., Ca, Mg, Zn, P) point to the potential of laser spectroscopy and spectrometry to identify tumor tissue.
