[eng][cze] This dissertation explores the regulatory mechanisms of Forkhead Box O (FOXO) transcription factors, with a focus on the interaction between FOXO4 and the tumor suppressor protein p53, which has significant implications for cellular senescence. FOXO proteins are an evolutionarily conserved family of transcription factors known for their critical roles in diverse cellular processes, including apoptosis, cell cycle arrest, and stress response. p53 is a homotetrameric transcription factor that controls many cellular processes, including metabolic adaptation, DNA repair, cell cycle regulation, and apoptosis. FOXO4 and p53 are key players in cellular senescence, a complex biological process marked by the irreversible arrest of the cell cycle. Studying the interaction between FOXO4 and p53 is crucial, as senescence is closely related to aging and age-related diseases. This study employs various biophysical techniques, particularly nuclear magnetic resonance (NMR) titrations, to identify the precise protein domains responsible for the interaction between FOXO4 and p53. The investigation extends to the structural characterization of the FOXO4:p53 complex using distance constraints derived from paramagnetic relaxation enhancement (PRE) NMR experiments. These experimental approaches provide a comprehensive...Tato disertační práce zkoumá regulační mechanismy transkripčních faktorů FOXO (Forkhead Box O) se zaměřením na interakci mezi FOXO4 a tumorovým supresorem p53, která má zásadní vliv na regulaci buněčné senescence. Proteiny FOXO jsou evolučně konzervovaná rodina transkripčních faktorů hrající klíčovou roli v různých buněčných procesech, jako je apoptóza, regulace buněčného cyklu a odpověď na stres. p53 je homotetramerní transkripční faktor, který řídí mnoho buněčných procesů, včetně metabolické adaptace, oprav DNA, regulace buněčného cyklu a apoptózy. FOXO4 a p53 jsou důležitými regulátory senescence, komplexního biologického jevu charakterizovaného nevratným zastavením buněčného cyklu. Studium interakce FOXO4 s p53 je zvláště důležité, neboť senescence je proces úzce spojený se stárnutím a nemocemi podmíněnými věkem.
