[cze][eng] Tato diplomová práce se zabývá speciační analýzou rtuti v lidských vlasech a nehtech pomocí metody postkolonového UV-fotochemického generování těkavých sloučenin (UV- PVG) v kombinaci s vysokoúčinnou kapalinovou chromatografií (HPLC) a detekcí pomocí atomové fluorescenční (AFS) a atomové absorpční spektrometrie (AAS). Cílem bylo optimalizovat podmínky přípravy biologických vzorků a experimentálních parametrů pro spolehlivé stanovení anorganické rtuti (Hgš⁺) a methylrtuti (MetHg). Byly porovnány různé metody extrakce, ověřen vliv pH, teploty a časové stability vzorků, stejně jako účinek chelatačních činidel. Výsledky ukazují, že použití AFS vykazuje nižší meze detekce a stanovitelnosti oproti AAS, a je tedy vhodnější pro stopovou analýzu. Metoda byla úspěšně ověřena pomocí referenčního materiálu lidských vlasů s obsahem rtuti, což potvrzuje její použitelnost při hodnocení expozice organismu rtuti prostřednictvím analýzy vlasů.This work deals with the analysis of mercury speciation in human hair and nails using the post-column UV photochemical generation of volatile compounds (UV-PVG) method in combination with high-performance liquid chromatography (HPLC) and detection by atomic fluorescence (AFS) and atomic absorption spectrometry (AAS). The aim was to optimize the conditions for the preparation of biological samples and the experimental parameters for the reliable determination of inorganic mercury (Hgš⁺) and methylmercury (MetHg). Various extraction methods were compared and the influence of pH, temperature, and sample stability over time, as well as the effect of chelating agents, were verified. The results show that using AFS has lower detection and quantification limits than AAS and is therefore more suitable for trace analysis. The method was successfully verified using reference material of human hair containing mercury, confirming its applicability in assessing mercury exposure through hair analysis.
