[cze][eng] Gramnegativní bakterie Bordetella pertussis je lidským patogenem způsobujícím respirační onemocnění zvané černý kašel (pertuse). K regulaci většiny faktorů virulence využívá tento patogen dvoukomponentní systém signální transdukce BvgAS, který je zásadní pro jeho virulenci. Systém BvgAS zajišťuje expresi širokého spektra genů jako adaptační odpověď na měnící se vnější podmínky během infekce. Tento systém je založený na dvou klíčových proteinech: transmembránové kináze BvgS a cytosolickém regulačním proteinu BvgA. Vnější podmínky ovlivňují aktivitu BvgS, která po stimulaci přenáší fosfát na BvgA protein, jenž poté funguje jako transkripční faktor. Na základě množství fosforylovaného BvgA lze rozlišovat tři fenotypové fáze bakterií B. pertussis, přičemž každý z těchto stavů je adaptován na konkrétní části životního cyklu bakterie. Vysoká hladina fosforylovaného BvgA odpovídá virulentní Bvg+ fázi, během které jsou pozitivně regulovány geny asociované s virulencí. Inaktivace aktivity BvgS kinázy indukuje avirulentní Bvg- fázi, která umožňuje přežívání v nepříznivém prostředí. Přechodnou fází mezi těmito dvěma stavy je intermediátní Bvgi fáze. Přestože přesné mechanismy regulace BvgAS systému in vivo nejsou objasněny, v in vitro podmínkách lze tento systém ovlivňovat tzv. modulátory, nikotinátem a...Bordetella pertussis is a Gram-negative bacterium and strictly human pathogen that causes whooping cough. Most of its virulence factors are regulated by the two-component signal transduction system BvgAS, which is central to its pathogenicity. BvgAS controls gene expression as an adaptive response to environmental changes during infection. BvgAS consists of the transmembrane sensor kinase BvgS and the cytosolic response regulator BvgA. Environmental stimuli modulate the activity of BvgS, which transfers a phosphate group to BvgA upon activation. The phosphorylated BvgA (BvgA~P) acts as a transcription activator. Depending on the intracellular concentration of BvgA~P, B. pertussis switches between three different phenotypic phases, each of which is adapted to a specific phase of its life cycle. High concentrations of BvgA~P lead to the virulent Bvg+ phase with active expression of virulence genes. On the other hand, when the kinase activity of BvgS is suppressed, the bacterium enters avirulent Bvg- phase, which promotes survival under adverse environmental conditions. An intermediate state, the Bvgi phase, represents a transitional phenotype between these two extremes.
