[cze][eng] Epiteliální karcinomy vaječníků tvoří nejčastější skupinu maligních karcinomů vaječníků a představují heterogenní skupinu onemocnění lišících se svým molekulárním profilem, histopatologickým vzhledem i klinickým chováním. Cílem této práce bylo analyzovat molekulární a histopatologické charakteristiky hlavních subtypů, kterými jsou high grade serózní karcinom (HGSOC), low grade serózní karcinom (LGSOC), světlobuněčný (CCOC), endometroidní (ENOC) a mucinózní (MOC). Dále je diskutováno využití moderních omics technologií (genomika, transkriptomika, epigenetika, metabolika, proteomika) při identifikaci molekulárních profilů nádorů a jejich význam pro vývoj cílené terapie. Pozornost byla věnována především typům genetických změn, kterými jsou variace počtu kopií (CNV) a jednonukleotidové varianty (SNV) a jejich vlivu na aktivaci nebo inhibici specifických signálních drah (TP53, MAPK/RAS, PI3K/AKT/mTOR, Wnt/β-katenin). Popsány jsou i rozdíly v chování jednotlivých subtypů. Závěrečná část poukazuje na doporučení pro další výzkum a na identifikaci neprozkoumaných témat. Klíčová slova: karcinom ovaria, molekulární onkologie, molekulární profilování, genomika, transkriptomikaEpithelial ovarian carcinomas constitute the most common group of malignant ovarian tumors and represent a heterogeneous group of diseases differing in their molecular profiles, histopathological appearance, and clinical behavior. The aim of this thesis was to analyze the molecular and histopathological characteristics of the main subtypes, which are high-grade serous carcinoma (HGSOC), low-grade serous carcinoma (LGSOC), clear cell carcinoma (CCOC), endometrioid carcinoma (ENOC), and mucinous carcinoma (MOC). Furthermore, the use of modern omics technologies (genomics, transcriptomics, epigenetics, metabolomics, proteomics) is discussed in the context of identifying molecular profiles of tumors and their significance for the development of targeted therapy. Attention was given primarily on the types of genetic changes, which are copy number variations (CNV) and single nucleotide variants (SNV), and their impact on the activation or inhibition of specific signaling pathways (TP53, MAPK/RAS, PI3K/AKT/mTOR, Wnt/β-catenin). Differences in the behavior of individual subtypes are also described. The concluding section highlights recommendations for future research and identifies areas that remain unexplored.
