Plicní fáze schistosomózy představuje klíčový moment interakce mezi migrujícími schistosomulami a imunitním systémem hostitele. Tato diplomová práce porovnává infekce způsobené ptačí schistosomou Trichobilharzia szidati a lidskou schistosomou Schistosoma mansoni u dvou inbredních kmenů myší (C57BL/6 a BALB/c) s cílem odhalit rozdíly v migraci parazitů, rozsahu tkáňové patologie a charakteru imunitní odpovědi. Zatímco T. szidati migruje parenchymem plicní tkáně a vyvolává časnou a výraznou zánětlivou reakci, S. mansoni migruje intravaskulárně, ačkoliv některá schistosomula skončí v alveolech, odkud pravděpodobně nejsou schopna zpětně proniknout do cévního řečiště a dochází k lokální infiltraci leukocytů. Schistosomula T. szidati byla u C57BL/6 deformována dříve, u některých s přítomností leukocytů na povrchu i uvnitř střevního lumen, a byla postupně eliminována, zatímco u BALB/c přežívala déle a déle přijímala potravu. Analýza cytokinového profilu (IL-4, IL-5, TNF-α, IFN-γ) a imunofenotypizace leukocytů v bronchoalveolární laváži (BAL) a plicní tkáni odhalila intenzivnější a déletrvající zánětlivou odpověď u C57BL/6 infikovaných T. szidati, kde zvýšené množství leukocytů přetrvávalo až do 42. dne po infekci. Histologické snímky však prokázaly přítomnost zánětu i u BALB/c, byť s odlišným charakterem... The pulmonary phase of schistosomiasis represents a critical stage of interaction between migrating schistosomula and the host immune system. This thesis compares infections caused by the avian schistosome Trichobilharzia szidati and the human schistosome Schistosoma mansoni in two inbred mouse strains (C57BL/6 and BALB/c), aiming to reveal differences in parasite migration, tissue pathology, and the nature of the immune response. While T. szidati migrates through the lung parenchyma and elicits an early and pronounced inflammatory response, S. mansoni migrates intravascularly, although some schistosomula become trapped in alveoli, from where they likely cannot return to the bloodstream, resulting in local leukocyte infiltration. T. szidati schistosomula in C57BL/6 mice exhibited earlier morphological deformation, often accompanied by leukocytes on their surface and within the intestinal lumen, and were progressively eliminated. In contrast, in BALB/c mice, schistosomula survived longer, showed delayed deformation, and continued to ingest host material. Cytokine profiling (IL-4, IL-5, TNF-α, IFN-γ) and leukocyte immunophenotyping in bronchoalveolar lavage (BAL) and lung tissue revealed a stronger and more prolonged inflammatory response in C57BL/6 mice infected with T. szidati, with elevated...
