[cze][eng] Tato bakalářská práce je zaměřena na problematiku ochucených sladových nápojů a představuje proces výroby sladu, jeho složení a význam jednotlivých technologických kroků, včetně máčení, klíčení a hvozdění. Dále je práce zaměřena na stanovení obsahu celkových antokyanů, rutinu a kyseliny askorbové v ovocných šťávách, sirupech a sladových nápojích připravených z černého bezu (Sambucus nigra L.) a černého rybízu (Ribes nigrum L.). V prvním kroku byl stanoven celkový obsah antokyanů s využitím spektrofotometrické metody. Později byla vypracována kalibrační řada pro obsah rutinu a kyseliny askorbové. Dále byl změřen obsah rutinu pomocí kapalinové chromatografie (HPLC) a na závěr, byl stejnou metodou stanoven obsah kyseliny askorbové. Tyto metody byly použity jak ve vyhodnocení obsahu zmíněných látek v ovoci, tak v sirupech z nich připravených, tak v ochucených sladových nápojích. V případě černého rybízu byl měřen obsah kyseliny askorbové a celkových antokyanů, rutin černý rybíz neobsahuje. Černý bez a jeho plody byly analyzovány na všechny zmíněné bioaktivní látky, zatímco květ černého bezu byl analyzován pouze na rutin. V závěru se práce zaměřuje na přípravu sladového nápoje ochuceného extrakty z černého bezu a černého rybízu a sleduje změny obsahu vybraných látek během jeho výroby. Cílem je zhodnotit možnost obohacení sladových nápojů o biologicky aktivní látky.This bachelor thesis focuses on the issue of flavoured malt beverages and presents the process of malt production, its composition and the importance of individual technological steps, including steeping, germination and mashing. Furthermore, the thesis focuses on the determination of the content of total anthocyanins, rutin and ascorbic acid in fruit juices, syrups and malt beverages prepared from elderberry (Sambucus nigra L.) and blackcurrant (Ribes nigrum L.). In a first step, the total anthocyanin content was determined using a spectrophotometric method. Later, a calibration series for rutin and ascorbic acid content was developed. Next, the rutin content was measured by liquid chromatography (HPLC) and finally, the ascorbic acid content was determined by the same method. These methods were used in the evaluation of the content of the above mentioned substances in the fruits, in syrups prepared from them and in flavoured malt beverages. In the case of blackcurrants, ascorbic acid and total anthocyanins were measured; rutin is not present in blackcurrants. Elderflower and its fruits were analysed for all the bioactive substances mentioned, while elderflower was analysed only for rutin. In conclusion, the work focuses on the preparation of a malt beverage flavoured with elderberry and blackcurrant extracts and monitors the changes in the content of selected substances during its production. The aim is to evaluate the possibility of enriching malt beverages with biologically active substances.
