[cze][eng] Diplomová práce se zabývá vlivem střevního mikrobiomu na regulaci chuti k jídlu u myšího modelu mentální anorexie založené na aktivitě a omezeném příjmu potravy (ABA - activity- based anorexia). Mezi poruchami příjmu potravy je mentální anorexie psychickým onemocněním s nejvyšší úmrtností, což zdůrazňuje její význam v hledání nových přístupů v její léčbě. V rámci experimentální studie byly použity myši s konvenčním, tj. nedefinovaným střevním mikrobiomem, kterým byla v průběhu prvních deseti dnů podávána antibiotika v rámci příprav střev pro kolonizaci lidským mikrobiomem transplantovaným ze zdravých kontrol a z pacientek s akutní nebo chronickou formou mentální anorexie. Cílem této práce bylo zjistit, zda transplantace střevního mikrobiomu z pacientek či zdravých kontrol ovlivňuje expresi hypotalamických peptidů regulujících chuť k jídlu a střevní permeabilitu. Výsledky práce ukazují, že fekální mikrobiální transplantací z pacientek a ze zdravých kontrol se složení myšího mikrobiomu výrazně přiblížilo dárcovskému složení. Významné změny byly též pozorovány i v rozdílu hmotností a aktivitě mezi myšmi s mikrobiotou transplantovanou od zdravých kontrol a také od pacientek s akutní či chronickou formou mentální anorexie. Na úrovni exprese proteinů orexigenních a anorexigenních byly zaznamenány výrazné...This thesis focuses on the influence of the gut microbiome in appetite regulation using a mouse model of activity-based anorexia nervosa (ABA). Among eating disorders, anorexia nervosa is the mental illness with the highest mortality rate, highlighting its importance in the search for new approaches in its treatment. In an experimental study, mice with a conventional, i.e., undefined, gut microbiome were administered antibiotics during the first ten days to prepare the gut for colonization with a human microbiome transplanted from healthy controls and from patients with acute or chronic forms of anorexia nervosa. The aim of this study was to determine whether transplantation of the gut microbiome from patients or healthy controls affects the expression of hypothalamic peptides regulating appetite and intestinal permeability. The results of this study show that faecal microbial transplantation from patients and healthy controls significantly brought the composition of the murine microbiome closer to that of the donor. Significant changes were also observed in weight and activity differences between mice with microbiota transplanted from healthy controls same as for patients with acute or chronic forms of anorexia nervosa.
