The Venom allergen-like (VAL) protein family is a diverse group of proteins with conserved structural elements. These proteins are ubiquitously present in excretory-secretory products of various helminths and are often associated with host invasion, tissue migration, and immune modulation. Schistosoma mansoni VAL family (SmVALs) has been characterized before in a great detail but suffers from outdated annotations and long-held assumptions about signal peptide distribution among its members and associated putative secretion. This thesis provides a comprehensive revision of the SmVAL gene family, identifying six previously undescribed genes, updating phylogenetic relationships, and providing new insights into the secretion potential of these proteins. Additionally, through refined meta- analysis of publicly available RNAseq datasets, we confirm that SmVAL gene expression is highly stage- and tissue-specific and add a valuable context to the egg - a life stage that has been largely overlooked in this regard. Using newly generated robust egg transcriptomic data from different tissues and developmental time points, we identified SmVAL9 and SmVAL29 as the most abundantly expressed SmVALs in mature eggs. Rodina Venom alergen-like proteinů (VAL) je rozmanitá skupina proteinů s konzervovanými strukturními prvky. Tyto proteiny jsou často zastoupeny v exkrečně-sekrečních produktech různých helmintů a jsou často spojovány s invazí do hostitele, migrací jeho tkáněmi a imunomodulací. Rodina VAL proteinů parazita Schistosoma mansoni (SmVAL) byla již dříve podrobně charakterizována, ale trpí zastaralými anotacemi a dlouholetými předpoklady o distribuci signálních peptidů a související domnělou sekrecí jejích členů. Tato práce poskytuje komplexní revizi genů kódujících SmVAL proteiny, vedoucí k identifikaci šesti dříve nepopsaných genů, aktualizaci fylogenetických vztahů a poskytuje nové poznatky o sekrečním potenciálu těchto proteinů. Kromě toho prostřednictvím zpřesněné metaanalýzy veřejně dostupných RNA-Seq datasetů v této práci potvrzujeme, že exprese SmVAL genů je vysoce specifická pro jednotlivá stadia a tkáně, a přidáváme cenný kontext pro stádium vajíčka, které bylo v této souvislosti do značné míry přehlíženo. Na základě našich nových robustních transkriptomických dat z vajíček různého stáří a izolovaných z různých tkání hostitele jsme identifikovali SmVAL9 a SmVAL29 jako nejhojněji exprimované SmVAL geny ve zralých vajíčkách.
