Ledviny jsou párovým orgánem plnícím řadu životně důležitých funkcí: udržují stálé vnitřní prostředí, stálou koncentraci minerálů, kyselost vnitřního prostředí, udržují objem celkové vody v organismu, ale vylučují také tři důležité hormony: renin, erytropoetin a kalcitriol. Udržováním vyrovnané koncentrace minerálů, především sodíku, a tím příslušného objemu vody v těle hrají ledviny velmi významnou roli v dlouhodobé regulaci krevního tlaku. Mechanismy ovlivňující regulaci krevního tlaku byly a jsou stále intenzivně studovány v mnoha laboratořích. Koncept navržený profesorem Guytonem, tzv. Guytonova teorie, se stal v renální fyziologii široce uznávaným a na tuto teorii navazuje řada nových poznatků prohlubující znalosti a chápání fyziologie ledvin a jejich výsadního postavení v regulaci krevního tlaku a rozvoji hypertenze (Guyton 1991, Hall et al. 1996, Navar 1997). Hlavním cílem této práce bylo zjistit, jakou úlohu hrají hlavní vazoaktivní systémy a jejich vzájemné interakce v regulaci renálních funkcí, jež by se významně podílely na rozvoji hypertenze. Přestože existuje celá řada vazoaktivních systémů, které ovlivňují renální funkce a krevní tlak a přímo se zapojují do jejich regulace, byly pro tuto práci zvoleny dva významné vazoaktivní systémy, a to systém oxidu dusnatého a renin-angiotenzinový systém.... The kidneys are paired organs that perform a number of vital functions: they maintain a stable internal environment, constant mineral concentration, and proper acidity of the internal milieu. They also regulate the total water volume in the body and secrete three important hormones: renin, erythropoietin, and calcitriol. By maintaining a balanced concentration of minerals-particularly sodium-and thus the appropriate volume of water in the body, the kidneys play a very significant role in the long- term regulation of blood pressure. The mechanisms influencing blood pressure regulation have been, and continue to be, intensively studied in many laboratories. The concept proposed by Professor Guyton, known as Guyton's theory, has become widely accepted in renal physiology, and many new findings that deepen our knowledge and understanding of kidney physiology and their prominent role in blood pressure regulation and the development of hypertension are based on this theory (Guyton 1991, Hall et al. 1996, Navar 1997). The main objective of this work was to determine the role of the major vasoactive systems and their interactions in regulating renal functions that may significantly contribute to the development of hypertension.
