Dědičné periferní neuropatie představují heterogenní skupinu geneticky podmíněných onemocnění, která významně ovlivňuje funkci periferního nervového systému. Hlavními klinickými projevy jsou pomalu progredující svalová slabost, atrofie a poruchy čití převážně u dolních končetin. Diagnostika je často velmi náročná, a to kvůli velké variabilitě symptomů a genetických příčin, kterými jsou mutace v genech kódujících strukturu a funkci periferních nervů. Moderní metody sekvenování nové generace výrazně rozšířily a urychlily možnosti genetické diagnostiky a dnes představují běžný přístup v rutinní diagnostice. Výzvu při používání této metody představuje zvyšující se objem sekvenačních dat, kvůli němuž roste potřeba jejich efektivního zpracování a ukládání, což vede i k využití cloudových technologií. Tato práce poskytuje přehled genetických příčin dědičných periferních neuropatií, přínos sekvenování nové generace v jejich diagnostice a možností využití cloudových platforem pro analýzu a správu genetických dat. Klíčová slova: dědičné periferní neuropatie, genetická diagnostika, NGS, cloudové technologie Hereditary peripheral neuropathies represent a heterogeneous group of genetically determined diseases that significantly affect the function of the peripheral nervous system. The main clinical manifestations are slowly progressive muscle weakness, atrophy and impaired sensation mainly in the lower limbs. Diagnosis is often very challenging due to the wide variability of symptoms and genetic causes, which are mutations in genes encoding the structure and function of peripheral nerves. Modern next-generation sequencing methods have greatly expanded and accelerated the possibilities of genetic diagnosis and are now a common approach in routine diagnosis. A challenge in using this method is the increasing volume of sequencing data, which has led to a growing need for efficient processing and storage, including the use of cloud-based technologies. This bachelorˈs thesis provides an overview of the genetic causes of hereditary peripheral neuropathies, the contribution of next-generation sequencing in their diagnosis, and the potential use of cloud platforms for genetic data analysis and management.
