Konzervační média určená k ochraně NK před degradací nám umožňují stabilizovat DNA po dobu několika let při pokojové teplotě bez množstevních ztrát nebo narušení její integrity. V některých institucích ale stále přetrvává přesvědčení, že zmražení je nejefektivnějším způsobem skladování DNA. Stejné nebo dokonce i signifikantně vyšší kvality a množství můžeme dosáhnout za určitých podmínek i s použitím jiných metod. Tato bakalářská práce diskutuje jedny z prvních stabilizačních procesů používaných při mumifikacích ve starověkém Egyptě společně se zkoumáním starobylé (ancient) DNA a oborů bioarcheologie zabývajících se analýzou koprolitů a jantaru. Diskutovány jsou i podmínky, které přispívají k degradaci molekuly DNA (teplota, světelné záření, vlhkost, pH, mikrobiální degradace) s konkrétními příklady vlivů způsobujících poškození genetické informace. Hlavní náplní práce je porovnání jednotlivých přístupů při stabilizaci DNA s důrazem na: rozdíl při uchovávání molekuly DNA v ne/přítomnosti stabilizačních činidel, skladování vzorku při pokojové teplotě nebo ve zmražené podobě, a vhodnost metody pro krátkodobou či dlouhodobou konzervaci. V práci jsou dále zmíněny přístupy vedoucí k optimalizaci výtěžku a kvality DNA s důrazem na jednotlivé druhy skladovaného biologického materiálu a detailněji jsou... Preservation media designed to protect DNA from degradation allow us to stabilize DNA for several years at room temperature without quantitative loss or compromising its integrity. In some institutions, however, the belief still persists that freezing is the most efficient way to store DNA. The same or even significantly higher quality and quantity can be archived under certain conditions using other methods. This bachelor thesis discusses some of the earliest stabilization processes used in mummification in ancient Egypt, along with the study of ancient DNA and the fields of bioarcheology concerned with the analysis of coprolites and amber. Conditions that contribute to the degradation of the DNA molecule (temperature, light, humidity, pH, microbial degradation) are also discussed, with specific examples of the effects that cause damage to genetic information. The main focus of the work is a comparison of different approaches to DNA stabilization, with emphasis on: the difference in the preservation of the DNA molecule in the absence of stabilizing agents, storage of the sample at room temperature or in frozen form, and the suitability of the method for short-term or long-term preservation.
