[cze][eng] Tato bakalářská práce se zabývá vztahem mezi vybranými tyrosinkinázovými inhibitory (TKI) a střevní bakterií Escherichia coli, která je přirozenou součástí lidského mikrobiomu. Bylo testováno pět různých inhibitorů - lenvatinib, vandetanib, lapatinib, sunitinib a cabozantinib. Téma této bakalářské práce je velmi aktuální, protože propojuje dvě klíčové oblasti současného biomedicínského výzkumu, které v posledních letech získávají stále větší pozornost - moderní onkologickou léčbu a střevní mikrobiom. Tyrosinkinázové inhibitory představují důležitou skupinu cílených protinádorových léčiv. TKI inhibují specifické enzymy zvané tyrosinkinázy, které hrají zásadní roli v signálních drahách regulujících růst a dělení buněk. Stále více se ukazuje, že složení mikrobiomu může ovlivňovat nejen účinnost protinádorových léků, ale také výskyt vedlejších účinků, například průjmů. Přestože se TKI běžně používají v klinické praxi, jejich možné interakce se střevními bakteriemi - konkrétně s běžnými druhy jako Escherichia coli - dosud nejsou dostatečně prozkoumány. Studium těchto vztahů proto může přispět k lepšímu pochopení individuální odpovědi na léčbu a pomoci při optimalizaci onkologické terapie v budoucnosti. Z výsledků měření absorbance vyplývá, že některé tyrosinkinázové inhibitory mohou mít stimulační...This bachelor's thesis focuses on the relationship between selected tyrosine kinase inhibitors (TKIs) and the intestinal bacterium Escherichia coli, which is a natural component of the human microbiome. Five different inhibitors were tested: lenvatinib, vandetanib, lapatinib, sunitinib, and cabozantinib. The topic of this thesis is highly relevant, as it connects two key areas of current biomedical research that have been gaining increasing attention in recent years - modern cancer therapy and the gut microbiome. Tyrosine kinase inhibitors represent an important class of targeted anticancer drugs. They inhibit specific enzymes known as tyrosine kinases, which play a crucial role in signaling pathways that regulate cell growth and division. It is becoming increasingly evident that the composition of the microbiome may influence not only the effectiveness of anticancer drugs but also the occurrence of side effects, such as diarrhea. Although TKIs are commonly used in clinical practice, their potential interactions with intestinal bacteria - specifically common species such as Escherichia coli - have not yet been sufficiently explored. Studying these interactions may contribute to a better understanding of individual responses to treatment and help optimize cancer therapy in the future. The results...
