[cze][eng] Itakonát je metabolitem Krebsova cyklu, kterému je v poslední době věnována velká pozornost, zejména díky jeho imunomodulačním vlastnostem. Podílí se na regulaci zánětu primárně prostřednictvím metabolických změn v buňce. Tato práce shrnuje účinky tohoto metabolitu, konkrétně jeho úlohu v metabolismu makrofágů, u kterých byl doposud nejlépe popsán. Itakonát inhibuje enzym sukcinátdehydrogenázu (SDH), ovlivňuje expresi prozánětlivých cytokinů prostřednictvím signálních drah NRF2 a NF-κB, potlačuje glykolytickou aktivitu, priming a aktivaci inflamazomu a reguluje produkci interferonů typu I. Díky jeho širokým modulačním schopnostem je itakonát vhodným kandidátem pro léčbu rakoviny nebo některých autoimunitních a neurodegenerativních onemocnění. It contributes to the regulation of inflammation primarily through metabolic alterations within the cell. This thesis summarizes the effects of this metabolite, focusing on its role in macrophage metabolism, where its function has been most thoroughly described to date. Itaconate inhibits the enzyme succinate dehydrogenase (SDH), influences the expression of pro- inflammatory cytokines via the NRF2 and NF-κB signaling pathways, suppresses glycolytic activity, priming and activation of the inflammasome and regulates type I interferon production. Due to its broad modulatory capabilities, itaconate represents a promising candidate for the treatment of cancer, as well as for certain autoimmune and neurodegenerative diseases.
