Student: Monika Berková, Vedoucí diplomové práce: PharmDr. Marta Kučerová, Ph.D., Konzultant: PharmDr. Petr Šlechta, Ph.D. Univerzita Karlova, Farmaceutická fakulta v Hradci Králové, Katedra farmaceutické chemie a farmaceutické analýzy Teoretická část diplomové práce se zabývá vývojem poznatků o boronových kyselinách jakožto potenciálních léčivech rakoviny. První část je zaměřená na 20. století, počínaje syntézami boronových kyselin pro testování při záchytné neutronové terapii, přes jejich využití jako inhibitorů serinových proteas až po objev inhibitoru proteasomu - bortezomibu. V druhé části je popsáno testování bortezomibu až do jeho schválení FDA. Třetí část se věnuje vlivu bortezomibu na vývoj inhibitorů proteasomu druhé generace a zvýšení zájmu o boronové kyseliny jako o potenciální protinádorová léčiva. Experimentální část je zaměřena na testování in silico, syntézu a biologické testování in vitro série boronových kyselin odvozených od struktury nesteroidního antiandrogenu flutamidu. Jeho užívání k léčbě rakoviny prostaty se pojí s rizikem selhání jater. Záměrem výměny nitroskupiny flutamidu za boronovou kyselinu bylo snížení toxicity na zdravé buňky a zároveň zachování cytotoxického účinku na buňky nádorové.... Student: Monika Berková, Supervisor: PharmDr. Marta Kučerová, Ph.D., Consultant: PharmDr. Petr Šlechta, Ph.D. Charles University, Faculty of Pharmacy in Hradec Králové, Department of Pharmaceutical Chemistry and Pharmaceutical Analysis The theoretical part of the thesis deals with the development of knowledge about boronic acids as potential cancer drugs. The first part focuses on the 20th century, starting with the syntheses of boronic acids for testing in neutron capture therapy, through their use as inhibitors of serine proteases, until the discovery of the proteasome inhibitor - bortezomib. The second part describes the testing of bortezomib up to its FDA approval. The third part discusses not only the impact of bortezomib on the development of second-generation proteasome inhibitors but also the increasing interest in boronic acids as potential anticancer drugs. The experimental part focuses on the in silico testing, synthesis and in vitro bioassay of a series of boronic acids derived from the structure of the non-steroidal antiandrogen - flutamide.
