[cze][eng] Předmětem této diplomové práce bylo elektrochemické studium látky 9-nitroantracenu (9-NA) na elektrodě ze skelného uhlíku, nalezení optimálních podmínek pro stanovení této látky a následně výzkum interakcí 9-NA s DNA pomocí metod stejnosměrné voltametrie (DCV) a diferenční pulzní voltametrie (DPV). Voltametrické chování této látky bylo zkoumáno v prostředí směsi ethanolu a Brittonova- Robinsonova pufru (BR pufr) v objemovém poměru 4:6. Optimální prostředí pro stanovení 9-NA bylo zvoleno pH BR pufru o hodnotě 4,0. V tomto prostředí byly proměřeny koncentrační závislosti 9-NA v rozmezí 1·10-7 - 1·10-4 mol/L. Dále byly proměřeny autentické vzorky pitné vody a modelové vzorky písku a štěrku. V další části bylo zkoumáno, k jakým interakcím zkoumaného analytu dochází v přítomnosti DNA a jak naopak interaguje DNA v přítomnosti tohoto analytu. Následně byly z těchto měření vypočteny vazebné parametry vzniklého komplexu analyt-DNA. Klíčová slova: Nitrované polycyklické aromatické uhlovodíky 9-nitroantracen Voltametrie Detekce poškození DNA DNA biosenzory Elektroda ze skelného uhlíkuThe subject of this Thesis was the electrochemical study of 9-nitroanthracene (9-NA) on a glassy carbon electrode, finding the optimum conditions for determining this substance, and subsequently researching the interactions of 9-NA with DNA using direct current voltammetry (DCV) and differential pulse voltammetry (DPV) methods. The voltammetric behaviour of this substance was investigated in a mixture of ethanol and Britton-Robinson buffer (BR buffer) in a volume ratio of 4:6. The optimum medium for the determination of 9-NA was selected as a BR buffer pH of 4.0. In this medium, the concentration dependence of 9-NA was measured in the range of 1·10-7 - 1·10-4 mol/L. Authentic samples of drinking water and model samples of sand and gravel were also analyzed. In the next part, the interactions of the analyte under investigation in the presence of DNA and, conversely, the interactions of DNA in the presence of this analyte were investigated. Subsequently, the binding parameters of the resulting analyte-DNA complex were calculated from these measurements.
