Glioblastom je jednou z nejagresivnějších a nejsmrtelnějších forem rakoviny, která se vyznačuje rychlým průběhem a extrémně špatnými výsledky léčby. Tradičně se výzkum glioblastomu spoléhal na dvourozměrné (2D) buněčné kultury a in vivo modely, které však mají značná omezení, pokud jde o přesné replikování komplexního mikroprostředí nádoru. Nedávné technologické pokroky umožnily vývoj trojrozměrných (3D) buněčných modelů, které nabízejí nové příležitosti pro lepší pochopení biologie glioblastomu a zlepšení terapeutických testů. Tato práce se zaměřuje na výhody 3D modelů ve srovnání s konvenčními 2D kulturami a poskytuje podrobné srovnání různých typů 3D modelů glioblastomu. Jsou diskutovány různé metody výroby těchto modelů spolu s jejich příslušnými aplikacemi v preklinickém výzkumu. Zdůrazněním výhod a výzev každého přístupu si práce klade za cíl zdůraznit význam 3D modelování pro pokrok ve výzkumu glioblastomu a podporu vývoje účinnějších léčebných postupů. Traditionally, glioblastoma research has relied on two-dimensional (2D) cell cultures and in vivo models, both of which have significant limitations in accurately replicating the complex tumour microenvironment. Recent advances in technology have enabled the development of three-dimensional (3D) cell models, offering new opportunities to better understand glioblastoma biology and improve therapeutic testing. This thesis focuses on the advantages of 3D models compared to conventional 2D cultures, providing a detailed comparison of different types of 3D glioblastoma models. Various methods for producing these models are discussed, along with their respective applications in preclinical research. By highlighting the benefits and challenges of each approach, the work aims to emphasize the importance of 3D modelling in advancing glioblastoma research and supporting the development of more effective treatments.
